Using double beam spectrophotometer from chemical Lab The spectrum is chosen at VIS range, wavelength 400nm 700nm. The Spectrum shows that non linear. There is no absorbance at concentration 0 mM, while 0.25mM, 0.5mM, 0.75mM and 1mM absorbance 4.4, 4.8, 4.6 and 3.9, respectively. The peak absorbance is at at 550nm. The absorbance at wavelength 400nm is quite linear to concentration. The low sample preparation could be cause non linear problem. Chemical reaction for five days incubate before measurement is the other reason.
Using single beam spectrophotometer at Photonics Lab (HR 4000 Ocean Optics) There are two peaks absorbance, at 420nm and 610 nm. The peak of absorbance is sharp and small difference between the concentrations, except 1 mM. The procedure in measurement technique is still unclear. The intensive training on using spectrometer HR 4000 Ocean Optics can solve the problem.
