6 Determining the concentration of an external solution which is isotonic to the cell sap of potato Problem statement: What is the concentration of an external solution which is isotonic to the cell sap of a potato cell? Hypothesis: The higher the concentration of sucrose solution, the lighter the weights of potato strip. // The concentration of the solution which is isotonic to the cell sap of potato cells has no effect on the weight of potato cells. Variables: a) Manipulated variable: concentration of sucrose solution b) Responding variable: weight of potato strips c) Constant variable: surrounding temperature and time Materials and apparatus: Razor blade, cork borer, test tubes, forceps, ruler, 50 ml beaker, ceramic tile, freshly cut potatoes, distilled water, sucrose solution with concentration 0.1M, 0.2M, 0.3M, 0.4M, 0.5M and 0.6M. Procedure: 1. Prepare seven test tube and label it A, B, C, D, E, F and G. (P1) 2. Fill test tube A with distilled water. (P1, P4) 3. Fill the other test tube with the following solutions: Test tube B C D E F G Sucrose solution 0.1 0.2 0.3 0.4 0.5 0.6 (M) (P1, P4) 4. Use a cork borer to obtain 7 potato strips. Cut each strip to a length of 3 cm. (P1, P2) 5. Wipe dries each potato strip with some tissue paper. (P5) 6. Weigh and record the initial mass of each strip of potato. (P3) 7. Place each strip of potato in each test tube and leave it for 30 minutes. (P1, P2) 8. After 30 minutes, remove each strip from the test tube and wipe dry. (P2, P5) 9. Weigh and record the final mass of each strip of potato. (P3) 10. Record the result in a table and draw a graph of the percentage difference in weight against the concentration of the sucrose solution.
