Biakan sel Fibroblas Embrio Ayam (FEA) dipersiapkan dengan metode dari
Methods for Examining poultry Biologics and for Identifiying and quantifiying
Avian pathogens (1971).
Telur ayam berembrio (TAB) umur 10 hari, berasal dari induk yang tidak
divaksin dan bebas terhadap penyakit tetelo (ND), Pullorum dan berdasarkan
pemeriksaan serologic tidak ditemukan serum anti terhadap penyakit marek.
Embrio diambil secara aseptik,pisahkan kepala,organ dalam dan anggota tubuh
lainnya. kemudian badan embrio yg sudah tertinggal dipotong-potong sampai
halusk. Potongan jaringan dicuci beberapa kali dengan larutan PBS sampai
larutan tampak jernih. Untuk setiap embrio ditambahkan larutan tripsin 0.2%
sebanyak 5 mL. Campuran tripsin-jaringan ini diputar dengan pengaduk magnit
selama 15 menit pada suhu kamar. Sel-sel ditampung dalam satu tabung. Proses
tripsinasi ini diulang 3-4 kali,hingga yang tersisa hanya jaringan-jaringan yang
kasar. Semua sel-sel yang terekstraks diputar dengan kecepatan 1000 rpm
selama 15 menit. Endapan sel pada dasar tabung disuspensikan dengan 100 mL
medium, kemudian disaring. Kepekatan sel dihitung dengan hemositometer,
apabila terlalu pekat suspensi sel diencerkan dengan media hingga diperoleh
kepekatan sekitar 5 x 10 5 sel/mL. Sel-sel dalam media ini selanjutnya
ditumbuhkan didalam tabung Roux, dengan volume 75 mL untuk setiap tabung.
Untuk membuat biakan sekunder, lapisan sel tunggal yang terbentuk pada
dinding tabung dipanen dengan larutan tripsin-versen,dicuci dengan larutan PBS
2-3 kali dengan pemutaran 1000 rpm, masing-masing selama 15 menit.
Endapan sel diresuspensikan dengan media pertumbuhan dan selanjutnya
ditumbuhkan didalam beberapa peralatan :

SUMMARY
Mareks disease

MD
)
is a lymphomatous and neuropathic dis

Serological tests:
Antibodies to MDV develop within 12 weeks of infection and are commonly
recognised by the agar gel immunodiffusion test,

serotypes 1 and 3 or trivalent vaccines consisting
of serotypes 1, 2, and 3 are al

A. INTRODUCTION
Mareks disease (MD) (Davison & Nair, eds., 2004; Schat & Nair, 2008; Sharma,
1998) is a disease of domestic fowl (chickens) caused by a herpesvirus. Birds get infect
ed by inhalation of infected dust from the poultry houses, and following a complex life
cycle, the virus is shed from the feather follicle of infect
ed birds (Baigent & Davison, 2004). MD occurs at 34 weeks of age or older and is
most common between 12 and 30 weeks of age. MD is associated with several distinct
pathological syndromes, of which the lymphoproliferative syndromes are the most
frequent and are of most practical significance. In the classical form of the di
sease, characterised mainly by the involvement of nerves, mortality rarely exceeds 10
15% and can occur over a few weeks or many months. In the
acute form, in which there is usually lymphoma formation in the viscera,
a disease incidence of 1030% in the flock is not uncommon and outbreaks involving up
to 70% can occur. Mortality may increase rapidly over a few weeks and then cease, or
can continue at a steady or slowly falling rate for several months. Currently, the acute
form of the disease with extensive visceral lymphomas is most prevalent. In
its classical form, the most common clinical sign of MD is partial or complete paralysis of
the legs and wings. In the acute form, birds are often severely depressed and some may
die without showing signs of clinical disease. Non-neoplastic disease involving brain
pathology with vasogenic oedema resulting in transient paralysis is increasingly
recognised with MD induced by the more virulent strains. In the classical form, the
characteristic finding is enlargement of one or more peripheral nerves. Those most
commonly affected and easily seen at post-mortem are the brachial and sci
atic plexuses, coeliac plexus, abdominal vagus and intercostal nerves.
Affected nerves are often two or three times their normal thickness, the
normal cross-striated and glistening appearance is absent, and the nerve may appear
greyish or yellowish, and sometimes oedematous. Lymphomas are sometimes present
in the classical form of MD, most frequently as small, soft, grey tumours in the ovary, and
sometimes also in the lungs, kidneys, heart, liver and other tissues. Grey eye caused
by an iridocyclitis that renders the bird unable to accommodat
e the iris in response to light and causes a distorted pupil is common in older (1618
week) birds, and may be the only presenting sign. In the acute form, the typical finding is
widespread, diffuse lymphomatous involvement of the liver, gonads, spleen, kidneys,
lungs, proventriculus and heart. Sometimes lymphomas also arise in
the skin around the feather follicles and in the skeletal muscles. Affected birds usually
have enlarged peripheral nerves, as in the classical form. In younger birds, liver
enlargementis usually moderate in extent, but in adult birds the liver may be greatly
enlarged and the gross appearance identical to that seen in lymphoid leukosis
, from which the disease must be differentiated. Nerve lesions are often absent in adult
birds with MD. In both the classical and acute forms of MD, the disease starts as a
proliferation of lymphoid cells, which is progressive in some cases and regressive in
others. The peripheral nerves may be affected by proliferative, inflammatory or minor
infiltrative changes, which are termed type A, B, and C lesions, respectively. The A-type
lesions consist of infiltration by proliferating lymphoblasts, large, medium and
small lymphocytes, and macrophages, and appear to be neoplastic in nature. The B-type
lesion is characterised by interneuritic oedema,infiltration by mainly
small lymphocytes and plasma cells, and Schwann cell proliferation, and appears to be
inflammatory. The C-type lesion consists of a light scattering of ma
inly small lymphocytes, and is often seen in birds that show no gross lesions
or clinical signs. It is thought to be a regressive, inflammatory lesion. Demyelination
frequently occurs in nerves affected by the A- and B-type lesions, and is
responsible for the clinical paralysis. Lymphomas in the visceral organs and other
tissues are similar cytologically tothe lymphoproliferations in the A-type lesions in nerves.
Usually the lymphoid cells are of mixed types, often wi
th a preponderance of small and medium lymphocytes, but sometimes, particularly in
acute MD in adult birds, large lymphocytes and lymphoblasts may predominate. The
heterogeneous population of lymphoid cells in MD lymphomas, as seen in haematoxylinand-eosin-stained sections, or in impression smears of lymphomas stained by May
GrnwaldGiemsa, is an important feature in differentiating the disease from lym
phoid leukosis, in which the lymphomatous infiltrations are composed of uniform
lymphoblasts. Another important difference is that, in lymphoid leukosis, gross
lymphomas occur in the bursa of Fabricius, and the tumour has an intrafollicular origin
and pattern of proliferation.In MD, although the
bursa is sometimes involved in the ly
mphoproliferation, the tum
our is less apparent, diffuse and interfollicular in
location. Peripheral nerve lesions are not
a feature of lymphoid l
eukosis as they are in MD. The greatest difficulty
comes in distinguishing between lymphoid leukosis and forms
of MD sometimes seen in adult birds in which the
tumour is lymphoblastic with marked liver enlargem
ent and absence of nerve le
sions. If post-mortems are
conducted on several affected birds, a diagnosis can
usually be made based on gross lesions and histopathology.
However there are other specialised
techniques described. The expression
of a Meq biochemical marker has
been used to differentiate between MD tu
mours, latent MDV infections and
retrovirus-induced tumours (Schat &
Nair, 2008). The procedure may require specialised reagent
s and equipment and it may not be possible to carry
out these tests in laboratories wi
thout these facilities. Other tec
hniques, such as detection by immuno-

Nerve lesions and lymphomatous
proliferations induced by ce
rtain strains of reticuloendotheliosis virus are similar,
both grossly and microscopically, to t
hose present in MD. Although reticul
oendotheliosis virus is not common in
chicken flocks, it should be borne in mind as a possibl
e cause of lymphoid tumour
s; its recognition depends on
virological and serological tests on the flock. Reticul
oendotheliosis virus can also cause neoplastic disease in
turkeys, ducks, quail and other species.
Another retrovirus, designated lymphopr
oliferative disease virus (LPDV),
also causes lymphoproliferative
disease in turkeys. Although chi
cken flocks may be seropositive for
reticuloendotheliosis virus, neoplastic disease is rare. T
he main features in the di
fferential diagnosis of MD,
lymphoid leukosis and reticuloendotheliosis are shown in
Table 1. Peripheral neuropathy
is a syndrome that can
easily be confused with the neurological
lesions caused by MD virus (MDV). This is not very common but its
incidence may be increasing in some European flocks (Bacon
et al.,
2001). There are no recognised health risks
to humans working with MDV or the related herpesvirus of turkeys (HVT).

RINGKASAN
Penyakit Marek (MD) adalah penyakit limfomatous dan neuropatik unggas domestik
disebabkan oleh alphaherpesvirus.
Diagnosis dibuat pada tanda-tanda klinis dan lesi kotor atau mikroskopis. Ayam dapat
menjadi terus terinfeksi dengan virus MD (MDV) tanpa mengembangkan penyakit klinis.
Infeksi oleh MDV adalah terdeteksi oleh virus isolasi dan demonstrasi antigen virus atau
antibodi.

Lesi saraf dan proliferations limfomatous disebabkan oleh strain tertentu dari virus
reticuloendotheliosis serupa,
baik terlalu dan mikroskopis, mereka yang hadir di MD. Meskipun virus reticuloendotheliosis
tidak umum di

Virus Reticuleondotheliosis dapat menyebabkan beberapa sindrom yang berbeda. Sindrom
limfoma bursal yang paling mungkin terjadi pada
8% natrium klorida, yang mengandung MDV antiserum. Tips bulu kecil yang diambil dari

Polymerase chain reaction (PCR)

quantitate beban virus dalam jaringan (Baigent et al, 2005;. Bumstead et al, 1997;. Burgess
& Davison, 1999;
. Reddy et al, 2000) atau diferensial mendeteksi MDV dan HVT dalam darah atau bulu tips
(Baigent et al, 2005.;
Davidson & Borenshtain, 2002).

2. Tes serologis
Kehadiran antibodi terhadap MDV pada ayam non-divaksinasi dari sekitar 4 minggu usia
merupakan indikasi
infeksi. Sebelum usia itu, antibodi tersebut dapat mewakili transmisi maternal antibodi
melalui kuning telur dan
bukan bukti infeksi aktif.
Virus, antigen dan antisera dapat diperoleh dari pemasok komersial atau dari OIE Reference
Laboratories untuk
Penyakit Marek (lihat Tabel pada Bagian 3 Manual Terrestrial ini), tetapi reagen standar
internasional belum
telah diproduksi.
a) imunodifusi Agar gel
Tidak ada tes diresepkan untuk perdagangan, tetapi gel imunodifusi agar (AGID) tes
digunakan paling
umum untuk mendeteksi antibodi. Tes ini dilakukan dengan menggunakan slide kaca dilapisi
dengan 1% agar fosfat
buffered saline yang mengandung 8% natrium klorida. Sumur yang berdekatan diisi dengan
antigen atau serum dan ini
diinkubasi dalam suasana lembab pada suhu 37 C selama 24 jam untuk difusi berlangsung;
positif sera acara
Reaksi identitas dengan serum positif dikenal dan antigen. Antigen yang digunakan dalam tes

Sel kultur jaringan MDV terinfeksi atau ekstrak tips bulu, atau kulit yang mengandung

sel fibroblast embrio ayam. Bila efek sitopatik adalah konfluen, sel-sel yang terlepas dari
budaya
kapal dan tersuspensi dalam media kultur atau fosfat buffered saline tanpa tryptose kaldu
fosfat
(Kehadiran kaldu fosfat tryptose dapat menghasilkan garis precipitin non-spesifik) pada
konsentrasi sekitar

sel / ml. Suspensi ini kemudian membekukan-dicairkan tiga kali dan digunakan sebagai
antigen.
Prosedur pengujian
i)
Membuat larutan 1% dari Difco Bactoagar di 8% natrium klorida dengan berdiri adonan ke
dalam air mendidih
bath.
ii)
Entah slide mikroskop atau cawan Petri dapat digunakan dan agar dituangkan ke ketebalan 23 mm.
iii) Potong lubang di agar menggunakan template dengan pusat baik dan 6 sumur spasi pada
jarak yang sama di sekitar
pusat dengan baik. Diameter sumur harus sekitar 5,3 mm, dan sumur harus sekitar
2,4 mm terpisah. Sebuah template dengan pemotong tersedia secara komersial.
iv) antigen ditempatkan di tengah dengan baik dan antiserum standar ditempatkan dalam
sumur eksterior alternatif.
Sampel serum yang akan diuji ditempatkan di sisa tiga sumur sehingga. Garis kontinu

b) Tes lain
Tes-tes lain untuk antibodi MDV meliputi tes antibodi fluorescent langsung dan tidak
langsung. Menunjukkan ini
kemampuan serum tes untuk noda MDV plak dalam kultur sel (Silva et al, 1997;. Spencer &
Calnek, 1970).
Tes ini adalah kelompok spesifik dan lebih sensitif dibandingkan dengan pengujian AGID.
Sebuah tes virus netralisasi untuk
kemampuan serum untuk menetralkan properti pembentuk plak dari MDV sel-bebas juga
dapat digunakan. Namun,
tes ini lebih cocok untuk tujuan penelitian selain untuk penggunaan diagnostik rutin.
Enzyme-linked
tes immunosorbent (ELISA) untuk mendeteksi antibodi MDV tersedia (Cheng et al, 1984;.
Sharma,
1998; Zelnik et al., 2004). Untuk mempersiapkan antigen untuk ELISA, sumur piring
microtitre 96-well yang dilapisi
dengan sel MDV terinfeksi.
C. PERSYARATAN VAKSIN
1. Latar Belakang

Persyaratan untuk memproduksi vaksin yang diuraikan di bawah ini, dan dalam Bab 1.1.8
Prinsip vaksin hewan produksi, namun sumber lain harus dikonsultasikan untuk informasi

a) Karakteristik benih
i) Karakteristik biologis
Virus dari kelompok MDV diklasifikasikan dalam tiga serotipe - 1, 2, dan 3 - atas dasar
mereka keterkaitan antigenik.
Serotipe 1: Ini mencakup semua strain patogen virus, mulai dari strain yang sangat
virulen plus (misalnya 648A), sangat virulen (misalnya Md / 5, Md/11, Ala-8, RB-1B),

rhinotracheitis virus. SPF bebek ternak harus bebas dari adenovirus burung, reoviruses
burung, Chlamydia, virus bebek enteritis, jenis virus hepatitis bebek I dan II, virus influenza
tipe A, virus penyakit Newcastle, Pasteurella (Sekarang Riemerella) anatipestifer, REV, dan
infeksi Salmonella. Kebebasan dari infeksi lain mungkin juga diperlukan karena mereka

b) Metode pembuatan
i) Prosedur

Untuk vaksin terkait sel, budaya terinfeksi HVT produksi atau MDV benih-virus saham,
dalam sel- bentuk terkait, yang biasanya dua bagian luar benih induk. Budaya diinkubasi
selama 48 jam maka sel-sel yang terinfeksi dipanen dengan memperlakukan lembar sel dicuci

of Federal Regulations, USA (1989); Thornton, 1985) and in Chapter 1.1.9 Tests for sterility
and
freedom from contamination of biological materials.
Keselamatan
Ten doses of vaccine or a quantity of diluent equivalent to two doses of vaccine should be
inoculated
into separate groups of ten 1-day-old SPF chickens. No adverse reactions should occur during
a 21day observation period.
With cell-associated vaccine, care is necessary to avoid injury from ampoules that may
explode when
they are removed from liquid nitrogen. Eye protection must be worn.
Batch potency
The standard dose of each type of vaccine is 1000 PFU per chicken or egg. Virus content
assays are
conducted on batches of vaccine to ensure that the correct dose per bird will be achieved.
c) Requirements for authorisation
i)
Safety requirements
Target animal safety
The master seed virus should be shown to be non-pathogenic for chickens by inoculating ten
times the
field dose into 1-day-old SPF chickens of a strain susceptible to MD, to ensure that it does
not cause
gross lesions or significant microscopic lesions of MD by 120 days of age. It should be noted
that some
vaccine strains of MDV and HVT may produce minor and transient microscopic nerve
lesions.
Reversion-to-virulence for attenuated/live vaccines
No increase in virulence should occur during six serial passages of the vaccine strain in 1day-old SPF
MD-susceptible chickens. Ten times the field dose of vaccine is inoculated initially and then
passaged
by inoculation of heparinised blood at 57-day intervals, and tests for viraemia are run to
check that
virus is transferred at each passage. The birds receiving the final passage are kept for 120
days and
should be free from MD lesions. However, some strains such as Rispens, may cause some
mild MD
lesi. The important observation is that the virulence should not change. This is a difficult test
because the genetic resistance of the chickens fundamentally affects the apparent virulence of
the
virus, so does the type of inoculum. After successful completion of laboratory safety tests, the
safety of
the strain should be confirmed in extensive field trials.
ii)
Efficacy requirements
A test for duration of immunity is carried out on the seed virus only. Such immunity is
apparently
lifelong. Preservatives are not included in the vaccine or diluent. During use, reconstituted
vaccine must
be kept cool and cell-associated vaccine should be agitated to keep cells in suspension.
iii) Stability
Tests for stability are carried out on six representative batches of vaccine to show that titre is
maintained during the stated shelf life of the vaccine. These tests should be conducted under
the
conditions of storage of the vaccine. The lyophilised product should have a shelf life of 12
months when
stored at 28C. Manufacturers may double the virus content of the vaccine to compensate
for some
loss of titre during storage. Appropriate diluting fluids are provided for use with cellassociated and
freeze-dried vaccines. The stability of reconstituted vaccine over a 2-hour period should be
tested.
3. Vaccines based on biotechnology
a) Vaccines available and their advantages
Although genetically engineered recombinant vaccines have been developed (Reddy et al. ,
1996) and tested
in laboratory and field trials (Lee et al. , 2010), they are currently not in commercial use.
