STAGE2DRYSEASON
(MET2IR,2012DS)

Introduction

IRRI breeding programs generate fixed and stable lines each season that are identified from pedigree
nurseriesaswellasobservationalandreplicatedyieldtrials.Eventually,elitelinesareadvancedtomulti
environmenttesting(MET)conductedviabreedingnetworks.Thesenetworks,however,presentlyhave
limited geographical coverage and face challenges in terms of germplasm movement. Moreover, the
materialstestedthroughthesenetworksaregenerallyintheadvancedstageofvarietaldevelopment.An
exhaustiveMETsystemforearlygenerationbreedingproductsissofarlackingatIRRIandintheglobal
rice breeding community. For this reason, a new MET system is being established at IRRI beginning in
2011undertheGlobalRiceSciencePartnership(GRiSP),tobepilotedforirrigatedlowlandrice.

Goal
Toestablishasystematic,sequential,multistage,andmultienvironmenttesting(MET)systemfor
elitebreedinglines,managedthroughoneentity,inordertoimproveoverallbreedingefficiency.

Objectives
Toidentifyelitebreedinglineswithhighandstableyieldsandwideadaptationacrossatarget
populationofenvironmentsthatcanbethefuturemegavarieties;
Tobeabletoselectsuperiorbreedingmaterialsadaptedtooneormorespecificenvironment(s)and
agroecologies;
Todevelopanddeployvarietiesandbreedingproductstospecificmarketsegments;
Toexploitgenotype,environment,andgenotypexenvironment(GXE)interactioncontributionsto
varietalperformanceinreleasingbreedingproductstooneormoretargetenvironments;
Togenerateearlierfeedbacktobreedersontraitperformanceandidentifytraitpackagesneededfor
molecularbreeding;
Toimprovepartnershipswithpublic,NGO,andprivatesectorswhomayhaverolestoplayatcertain
testing/varietydevelopmentstages..

TrialComposition

The MET2IR trial is the second stage of testing for irrigated breeding lines from IRRI and collaborating
NARESinstitutionselectedpromisinglinesfromMETStage1forirrigatedbreedinglinesundertheoverall
METconcept.For2012dryseason,thetrialwillconsistof140testentriesand5checks.Thetestentries
willmostlyincludebreedinglinesdevelopedbythevariousbreedingprograms(bothhybridandinbred)of
IRRIandPhilRice.Thedesignation,sourceandotherinformationofeachentryareprovidedinTable1.

TheExperimentalSite

Anexperimentalfielduniforminsoiltexture,depth,andfertilityshouldbechosenfortheMET2IRtrial.It
shouldnothavebeenusedpreviouslyforfertilizerexperiments.Itshouldalsohaveadequateirrigation
anddrainagefacilities.Theareacovers0.5hectareoflandpersite.Itissuggestedthatthesameareabe
utilizedfortheMETeveryseason.

ExperimentalDesign

The trial will be conducted using a RowColumn Design with two replications. This design is used to
considerpossiblefieldvariation.Therearetwomodules,basedonthefloweringtimeofentriesModule
1forveryearly/earlyfloweringand,Module2formedium/lateflowering,asfollows:

Foreachreplicate,module1has30rowsand4columnswhilemodule2has15rowsand4columns.

*Thisfieldbookcontainstheplotrandomization,fieldplananddatasheetsfortheMET2IR2012fordry
season.


For each module, the test entries are randomly assigned plot numbers. Within each module, the 2
replicates have different randomizations. All five checks are randomly assigned in a replicate and are
included within each column (checks are systematically arranged on every 6th row to help on PACP
scoring).Differenttrialsetshavedifferentrandomizationofentries.Theplotrandomizationofentriesis
giveninTables2and3.Thereare120plotsforModule1whileModule2,has60plots.Theseedpackets
arearrangedandnumberedaccordingtoplotnumbers.

The field layout in Figure 1 is provided for your reference. Please do not modify the design or the
layout.

Preparethefieldthoroughlyatleastonemonthbeforetransplantingfollowingthelocallyrecommended
standards.Layoutrowsinbedsbeforeseeding.Theexperimentalfieldshouldbeproperlylabeled.The
plotsizeforeachentryis10m2=2.0mx5.0m(10rowswith20cmdistancebetweenrows).

Thereshouldbenovacantrowsbetweenplots(entries).Numbertheplotsconsecutivelyfromlefttoright
inallreplications.Placethestakesbearingtheplotnumberatthefirstleftmostrowofeachplot.Labelall
plotsbeforedistributingtheseedlingsfortransplanting.
Applymolluscicidetwotimes,i.e.onceatfourdaysbeforeandanotherimmediatelyaftertransplanting.

NurseryEstablishmentandManagement

Initiallyallseedsareplacedinanovenforbreakingdormancyat50oCfor5days.Atotalof100gramsof
seedsforeachentryareseeded,whichshouldallowtheplantingof2replicationseachwith2mwidth(10
rows)x5.0mlength(25hills).Theseedscouldbeseededineitherwetordrybedswheretheyaregrown
for 21 days. The seedling beds are carefully kept free from insect and diseases and properly irrigated.
Afterseeding,applyuniformly1kg/100m2ofAmmoniumSulfate(2100S)intheseedbed.Iftheseedlings
showyellowing(Ndeficiency),applyanother1kg/100m2ofAmmoniumSulfate(2100S)1012daysafter
seeding(DAS).

Priortopulling,preparetagstomarkseedlingstobetransportedfromseedbedtothefield.After21days
ofsowing,eachseedlingoftheentriesarepulledoutfromtheseedlingbeds,bundled,andtiedwithaG.I.
wiremarkedwithpotlabelsbearingtheplotnumbers.Distributeseedlingsinthefieldcorrespondingto
thelayout.Checktheentriesandtheplotsforanypossiblemistake(s)beforetransplantingtheseedlings.
Alternatively,apply zinc sulfate in thenursery seedbed,or dip seedlings in 24 % zinc oxide suspension
beforetransplanting.(Ifzincsulfateistobeappliedinthesoilasbasal,dippingisnolongernecessary.)

Theseedlingsaretransplantedintothefieldin23seedlingsperhillfollowingaspacingof20cmx20cm.
Keepsomeseedlingsattheendoftheplotneartheplotlabeltoreplantmissinghills.

Shallowwaterdepthshouldbemaintainedstartingfromabout3daysaftertransplantingthatisgradually
increasedto35cmuntiltheharddoughstage.Replantmissinghillswithin7daysaftertransplantingto
obtain a uniform plant population. Care should be taken to uniformly distribute fertilizers and plant
protectionchemicals.

For weed control, a preemergence herbicide (Sofit EC if available) and molluscicide may be applied
uniformly in the experimental field immediately after transplanting following the recommended rate.
Maintainashallowwaterdepthof25cmforatleast2weeksafterherbicideapplication.Subsequent
handweedingmaybedoneifneeded.

Fordiseaseandinsectcontrol,theexperimentalfieldshouldbewellprotected.Choosetheappropriate
cultural, chemical, and biological control to effectively manage insects and diseases. If available,
carbofuranmaybeappliedataround15DATandatPI.DoprophylacticapplicationofBenlateatPIand
followupspray2weekslaterforsheathblight.Rougingshouldbedonetoremoveofftypeplants.Rogue
thefieldrepeatedlyuptotheharddoughstageorforaslongasofftypescanbeidentified.Theseoff
typeplantsshouldbecutatthebase.

Atharvest,collectalltheplantsinaplotexcepttheborderrows,oneoneachsideoftheplotandtwo
borderrows,oneonbothendsofalltheplots(Thetotalnumberofplantsharvestedfromaplotis8rows
x 23 plants). The harvest from each plot is placed in clean cloth bags. Put the labels, bearing the entry
number and plot number inside the bag. A vogel thresher is used to thresh the samples. Clean the
thresher well after every sample. Put the label inside the bag after threshing. All threshed samples are
dried in a batch dryer at 45oC for 45 days. In removing half and empty grains, clean the seed blower
before processing each sample to avoid seed mixtures. Transfer clean seeds into labeled paper bags
bearingtheMETname,METentrynumber,plotnumber,year,andseason.

The general reference for data collection is the Standard Evaluation System for Rice (SES, 1996). The
