The principle of this analysis is as follows: When a solution of CsCl is centrifuged at high speed, a concentration gradient develops as the consequence of an equilibrium being reached between sedimentation and diffusion. The Cs-salt of DNA, if mixed into the centrifuge tube, will within a couple of days from a narrow band at the CsCl concentration that corresponds to its buoyant density. 14N-and 15N-containing DNA molecules have different buoyant densities and thus can be distinguished in CsCl density gradients. (Note that this centrifugation is different from sucrose gradient centrifugation, where rates of sedimentation are determined.)
Meselson and Stahl observed that, after transfer to the light medium, more and more of the DNA, which originally banded at the heavy 15N-position in the gradient, formed a new band at a position of lighter density until, after one generation, essentially all DNA was of a hybrid density in-between that of fully 15Nlabeled heavy DNA and that of light 14N-labeled DNA. In the course of a second round of replication (generation) in the light medium, a new band gradually appeared at the position of light DNA while the hybrid band remained unchanged in quantity (next figure).

El genoma de un organismo es el juego completo de ADN. La planificacin del Proyecto Genoma Humano se inici en 1986, previsto para el 2007.
