* Dehydrated powder, hygroscopic in nature, store in a dry place, in tightly-sealed containers below 25C and protect from direct Sunlight. Instructions for Use Dissolve 35.40gms in 1000ml of distilled water. Gently heat to boiling with gentle swirling and dissolve the medium completely. Sterilize by autoclaving at 15 psi (121C) for 15 minutes. Mix well. Cool to 45500C and aseptically add 5 - 10% v/v sterile defibrinated horse blood and dispense into sterile test tubes or vials as desired. Appearance: Light amber colour, clear to slightly opalescent gel. On addition of 5 - 10% v/v sterile defibrinated horse blood; cherry red opaque solution appears pH (at 25C): 6.8 0.2 Principle ANAEROBIC BASAL BROTH is used for cultivation of anaerobic microorganisms like Bacteroides and other fastidious anaerobes. Anaerobic diphtheroids are among the major inhabitants of the human oral cavity. Scientists reported indicated that most of the oral anaerobic and human skin diphtheroids were identical to Propionibacterium acnes in their biological and serological characteristics. Medium contains Peptone as a source of nitrogen, vitamins & minerals. Dextrose is a carbon energy source. Sodium thioglycollate provides a low oxidation-reduction potential. Sodium chloride helps to maintain the osmotic balance of the medium. Starch is helpful in absorbing toxic metabolities produced by colonies. Amount of arginine is added to ensure the growth of Eubacterium lentum. Hemin and Vitamin-K serves as growth factors for many Bacteroides species. Sodium succinate also improves the growth of Bacteroides species. Sodium pyruvate is the energy source and acts like catalase and degrades traces of hydrogen peroxide. L-cysteine hydrochloride and dithiothreitol act as reducing agents. Colonies were isolated randomly from the sample and, after a second purification, isolates were subjected to catalase production testing and gram staining. The biological characteristics of the gram-positive, catalase-positive rods were

examined according to the Anaerobe Manuals. Interpretation Cultural characteristics observed under anaerobic conditions with addition of 5% w/v sterile defibrinated blood, after inoculating (103CFU/ml), on incubating at 35 2C for 48 - 72 hours.
