How GigaMune Rep-Seq Works


1 Extract genomic DNA from tissue.
Optionally, GigaGen can extract DNA from tissue samples. Acceptable tissues include peripheral blood and organ biopsies. Pre-sorting by flow cytometry or magnetic beads is optional.

Amplify TCR variable regions.


Rearranged TCR variable regions are amplified from genomic DNA by multiplex PCR using a set of 20 primers to amplify across all 59 V segments and 13 primers to amplify across all 13 J segments.

Sequence TCR variable regions.


Sequencing tags are added to amplified TCR variable regions in preparation for ultra-deep high-throughput sequencing.

Process sequencer output.


A custom analysis pipeline processes sequencer output, identifying V and J segments, translating TCR variable regions, and quantifying frequencies of individual T cell clones.

Interpret, analyze, and visualize TCR repertoire data.


View data using ClonoByte web-based software for data storage, processing, statistical analysis, sample comparison, and data visualization in the cloud.
