. 1. Purpose To ensure proper operation of Autoclave and Sterilization of media/apparatus etc. 2. Scope This procedure is applicable to operate Autoclave available in Q.A. Lab. 3. Responsibilities Microbiologist AM-Quality. 4. Definition : SOP: Standard operating procedure. 5. Procedure 1) Remove the inner liner. Fill the outer chamber of the autoclave with water up to the marked height. 2) Place the articles to be sterilized into the inner liner. The container should not be packed tightly. It is important that steam circulates freely. Place the inner liner back into the autoclave. Sterilization check indicator (Started-line indicator) needs to be put inside the liner containing the load. 3) Check that the Lid Gasket is correctly sealed in its groove on the under side of the lid. Place the lid on the autoclave and rotate clockwise until it hits the slops and the slots for the drops Bolts line up in the lid and body ring. Now tighten up in opposite pairs. 4) Close the Draw-off Cock and open the Airlock. 5) Plug the power lead of autoclave into the mains (the green lights will show connection to the mains).Turn on the power Switch (the red light will show power on). 6) When the water begins to boil, steam will start to emit from the Airlock. It will splutter at first. Close the airlock once it has stopped spluttering and a clean jet of steam is visible. Ensure that a clean jet of Steam is visible before closing the Air-lock to expel all the air from the Autoclave. 7) The sterilization period begins are as follow. Instruments and Equipment Dressings and Drapes Liquid in sealed Containers(0ml-50m) Liquid in sealed Containers(50ml-100ml) Liquid in sealed Containers(100ml-200ml) Culture Media (up to 1 liter) Prepared By Microbiologist Approved By Management Representative

8) When the sterilization period is complete, turn off the power Switch. 9) When the autoclave is cooled, the airlock should be opened. Check that the Pressure Gauge is reading zero. Unscrew the Nuts in opposite pairs one in each hand 2 full turns. Twist the lid anticlockwise until the locking lugs disengage.
