BIOCHEMISTRY LABORATORY II BCHS 4311 Lecture Syllabus Spring 2014

Lab Coordinator: Dr. Donna Pattison Office: Room 108C, Science Teaching Lab Building Phone Number: 713-743-2642 e-mail: dpattison@uh.edu Text: Laboratory DNA Science; Bloom, Freyer, and Micklos (1996) [ISBN: 0-80533040-2] Lecture Time/Location: Mondays, 9:00-10:00 am; AH110 Office Hours: Monday/Wednesday 10-11 am and by appointment Course Overview and Objectives: Students will gain first-hand experience using some of the most common and useful tools employed by biochemists and molecular biologists today. By the end of the course students will understand and be able to set up a polymerase chain reaction, clone a gene into a vector, transform a cloning vector into a cell line, and purify expressed protein. Students will be able to explain when and why scientists would use these particular methods in the laboratory. Students will be able to follow the scientific method from start to finish. Students will develop their own problem to explore in the context of the biomaterials project. They will establish a hypothesis, design experimental methods, conduct the experiment, analyze the data, and communicate their findings to their peers in both oral and written form. Students will learn the basics of professional writing for the sciences and demonstrate mastery of the skill in two written manuscript papers during the course. Lecture Focus: In lecture we will discuss the why behind the steps in a protocol. Molecular biology protocols are not just recipes to be followed. Understanding the reason the steps are written will help you design your own experiments and protocols in the future and help you understand the discipline of molecular biology itself. We will also discuss the types of research and applications in which the techniques you will be learning are used. The elements of scientific writing will be covered in lecture extensively. Attendance and Grading Attendance at lecture and labs is MANDATORY. The introduction to the lab for the week will usually be given in lecture rather than in lab. Most of the questions on your lab quizzes and the final exam will be written by Dr. Pattison and will be based on material covered in lecture. Therefore, it is in your best interest to attend lecture. Attending labs alone will NOT prepare you for the quizzes. The grading policy can be found on the lab syllabus. Clickers Clickers will be used in lecture to track your attendance and your understanding of the material. Information on grading can be found in the lab syllabus. You may either use a clicker or use your WiFi enabled device to earn your clicker points. Information on obtaining, registering, and using your clickers or devices can be found at the end of the syllabus.

Students with Special Needs: The Americans with Disabilities Act of 1990 requires that universities make reasonable accommodations to persons with disabilities as defined in the act. Students who feel they need assistance as defined by the guidelines set forth in the act should contact the lab coordinator, Dr. Pattison, to discuss appropriate arrangements.

Clickers or Registration Codes
You may select from one of two response device options: 1. Student ResponseCards: Students may purchase or use a TurningPoint ResonseCard to participate in class sessions. You may purchase these from the bookstore for $40 (rental clickers are $20). You may also use devices purchased or borrowed from friends or other outlets (as long as your friend is not also registered for this class). The device pictured below is supported here at UH (NOTE: you can use either the LCD screen version shown below or the older version without the LCD screen):

2. ResponseWare: Students may also choose to participate using ResponseWare. ResponseWare can be accessed from mobile devices using the iOS (Apple), Android, Blackberry, or PalmOS platforms. At this time, there is no app for Windows devices. The app is free to download, but you still need to purchase a registration code based on the amount of time you will need the app. You can purchase these through the bookstore or directly through TurningPoint by going to https://store.turningtechnologies.com/and entering in the code gG9N when prompted for a school code. You can also use ResponseWare through any web browser by going to www.rwpoll.com.

How to register your clicker or device via Blackboard


Please follow the steps below to register your clicker: 1) Log on to Blackboard at www.uh.edu/blackboard. 2) Select the course you will be using the TurningPoint clicker for. Click on Tools, click the Turning Technologies Registration Tool, click Response Card. Do not attempt to register the ResponseWare account through the ResponseWare registration area. Whether you are using

a traditional clicker or your phone/tablet, etc., you will need to click the Response Card link!

Enter your clicker ID. The ID is available on the back of your clicker it is a 6-digit combination of numbers or/and letters below the bar code. There is no letter O or I in the ID. NOTE: If you already registered your clicker for a course in the previous or current semester, you may see the following statement when you try to register your clicker for another course. Even though it shows that your clicker is already registered, provide your clicker ID again.

How to use the clicker in class


1. Remove Plastic Flap: If you have a brand-new clicker, remove the plastic flap from the back of your clicker before using it. 2. Set the Channel for the course: (Note that each course will have a different channel number) To set the channel,

Press CH channel number CH (LCD Model) Press GO channel number GO (Non-LCD Model) If done correctly, you should see a green light on your clicker. IMPORTANT You need to set the channel EVERY time you come to class. 3. Submit Answer: After the channel is set at the beginning of the class, you only need to press your answer choice, such as A, B, C, D or 1, 2, 3, 4 to submit your response. NOTE: DO NOT press Go or Ch before pressing your answer option. Your response will not be received. 4. Check for the Green Light: When you submit an answer, pay attention to the light indicator. If the light is solid green, it means that your answer has been received. You can change your answer as long as the polling is still open. Only the last answer submitted will be recorded.

Commonly Asked Questions


What does a red light after submission mean?
A red light indicates that the response was not received. It could be because of any of these reasons: You pressed CH (or GO) to check the channel before submitting your response. Wait for the light to go off and then press ONLY your answer choice. You are on the wrong channel. Try resetting the channel and answering again. You responded before polling opened or after it closed.

What if I have to replace my clicker?


Register the new device in all your classes. Then, inform all instructors immediately of the change and provide them your old and new device IDs this is so points can be transferred from the old clicker to the new one.

What type of battery does the clicker require?


Each clicker comes equipped with two 3.0V lithium coin cell batteries model number CR2032. Replacements can be purchased at the campus bookstore and most electronic retail outlets. The campus bookstore sells clicker batteries for $2 for a pack of two. Check your battery indicator regularly and carry a spare set of batteries with you.

Where can I get help if my clicker fails?


In general, your clicker comes with a one-year warranty from the day of your clicker purchase. If your clicker stops working during the warranty period, bring your non-working clicker to Faculty Development and Instructional Support (FDIS).

If you encounter problems with your RF-LCD clicker (non-battery issues) even after the one-year warranty period has expired, you can still receive a replacement if it appears to be malfunctioning. If you have questions regarding your clicker points, please contact your instructor or TA.
