Summary
T e increasing pre!alence of dental caries is ma"ing it more of a major #orld ealt problem. Caries is t e direct result of acid production by cariogenic oral bacteria$ especially %treptococcus mutans. &e# and better antimicrobial agents acti!e against cariogenic bacteria are badly needed$ especially natural agents deri!ed directly from plants. 'e a!e e!aluated t e in ibitory actions of ()-mangostin$ a *ant one purified from et anolic e*tracts of t e tropical plant +arcinia mangostana L.$ by repeated silica gel c romatograp y. ()-Mangostin #as found to be a potent in ibitor of acid production by %., mutans -A./0$ acti!e against membrane en1ymes$ including t e 23H45-ATPase and t e p osp oenolpyru!ate, - sugar p osp otransferase system. ()-Mangostin also in ibited t e glycolytic en1ymes aldolase$ glyceralde yde-6-p osp ate de ydrogenase$ and lactic de ydrogenase. +lycolysis by intact cells in suspensions or biofilms #as in ibited by ()-mangostin at concentrations of .7 and .78, ,9mol,:L-.$ respecti!ely$ in a pH-dependent manner$ #it greater potency at lo#er pH !alues. ;t er targets for in ibition by ()-mangostin included 3i5 malolactic fermentation$ in!ol!ed in al"ali production from malate$ and 3ii5 &A<H o*idase$ t e major respiratory en1yme for %., mutans. T e o!erall conclusion is t at ()-mangostin is a multitarget in ibitor of mutans streptococci and may be useful as an anticaries agent.
