Reactive Oxygen Species (ROS) generated by oxidative stress caused due to various environmental factors and climatic conditions has been associated with human ailment and food deteriorations. Free radical mediated oxidative damage to biomolecules including protein and DNA has been complicated with cancer disease development. Role of antioxidants in combating oxidative stress related problem has been well established and search for novel anti-oxidant agents is one of the thrust areas of research in biomedical sciences, cosmetic and food industries. The current researches describe isolation and characterization of a natural potential antioxidant from Aegle marmelos leave. Boiling water extracts are usually being prepared from A. marmelos fresh leaves and extracts are further fractionated with liquidliquid partitioning method into n-hexane, diethyl ether, ethyl acetate, methanol, ethanol and water fractions. The GC-MS and LC-MS study indicated the presence of gallic acid, querecetin and ferulic acid as major antioxidant constituents. The results obtained from DPPH (2, 2-diphenyl-1-picrylhydrazyl) free radical scavenging activity assay revealed that only ethyl acetate and methanol fraction showed most of the scavenging activities with EC50 concentration 20 and 14 g per ml extract fraction, respectively. The deoxyribose degradation experiments are carried out using hydroxyl radical generated from iron mediated Fenton reaction. The results revealed significant hydroxyl radical scavenging potential with EC50 108 and 86 g per ml extract fraction for ethyl acetate and methanol fraction, respectively. agent. The protective potential against oxidative damage to calf thymus DNA and plasmid are studied on 1% agarose gel using copper (II) and ascorbate reaction mixture as source of free radical damage. The densitometric analysis of DNA band intensity are performed to study the extent of protection against control DNA damage. The results showed marked inhibition potential against oxidative damage to DNA with EC50 120 and 93 g per ml extract fraction for ethyl acetate and methanol fraction, respectively. The protective effect of ethyl acetate and methanol fraction on hypochlorite mediated oxidative damage to bovine serum protein are studied based on native PAGE and spectrophotometer technique. The results revealed that both fractions ethyl acetate and methanol showed significant inhibition potential with EC50 220 and 158 g per ml extract fraction, respectively. On the basis of above findings, scientists found these two fractions ethyl acetate and methanol as promising source of antioxidant activities and convincing protective role against oxidative damage to DNA and protein.
