Introduction the experiment is trying to examine how pH effect on urease enzyme activity and to test the function of urease enzyme in difference condition (pH, temperature or ionic condition) and observe how the function of the enzyme change in the certain condition. We predict that the pH of the solution will affect the rate of activity. Only the proper rate of pH will give the best result. Other than that, the enzyme will be denatured. Answer: To check if the enzyme still working when we add the soya suspension. 2. why did we need to add an enzyme in to a test tube without urea even though we can predict that no enzyme activity will occur? Answer: we need to add an enzyme into a test tube with out urea in order to see the different when the enzyme activity occur and proof that the enzyme is really work. Effect of pH on urease 1.How does pH effect on urease activity? How do you do? Answer: The urease enzyme function will work more efciently in certain pH that we can know by measure the pH and the change of Colour. 2.From your result, what pH would you use and avoid if you have to conduct an experiment using urease enzyme and why? Answer: According to the result, we would use the substance which is have the pH around 4 and avoid using substance which have the pH more than 7 because it will denature. Effect of pH on urease 3. Because basic pH will also change the colour of the indicator to blue/green which is very similar when ammonia has been produced by the urease enzyme, how can we make sure that the colour change is actually due to the urease activity? Is it possible we can make sure that the colour change is actually due to the urease activity by measure the pH twice (before and after add urea solution) in order to make the result of the experiment more accurate. Conclusion After the experiment, we have learnt that enzyme will work most efcient at the certain level of pH. If the pH level is too high or too low, enzyme can denature and make it unable to bind substrate into another shape. Based on our hypothesis which said that pH is the one factor that affect rate the activity that will occur. From the result, the enzyme was more likely able to work in more base condition. On the other hand, in acidic solution, the enzyme was denatured. The effect can be observed through each test tube. In tube that was in acidic conditions, there was a white solid curd under the tube. For the base solution, the colour that was changed too was same as the subjected solution from procedure 2. After we compare, it obviously shows that pH affect rate of activity. There is some error in the experiment which is we havent count the number of citric acid solution and sodium bicarbonate correctly. We just observe base on the colour of the solution.
