The existence of the cheese begins when a shepherd found the dairy cows kept in a bag of goat
leather, frozen and fermented when left in the cave. A hungry man to eat clumps of milk before and
liked the taste. Since then deliberately fermented milk to produce food that we later know as cheese.
Long ago the farmers keep the rest of the milk and let it clot. After that, the lump of cheese pounded

Making Cheese
Cheese making is the process undertaken to process the milk to become different kinds of
cheese. Making cheese is basically the same although there are hundreds of types of cheese are
produced worldwide. [1] Cheese has a style and taste different, depending on the type of milk used,
the type of bacteria or fungus used in fermentation, long process of fermentation and
storage ("maturation"). [1] Other factors such as type of food consumed by the mammal producing
milk and milk heating processes. [1] There are five main stages in the manufacture of cheese. [2]

menghasilkan energi yang besar. Di dalamnya terkandung kalsium dan phosphorus, terutama kejukeju yang dimasak. Keju sangat baik untuk menggantikan susu terutama bagi mereka yang tidak
menyukai susu.

Pembuatan keju
Pembuatan keju adalah proses yang dilakukan untuk mengolah susu hingga menjadi berbagai jenis
keju. Pembuatan keju pada dasarnya sama walaupun ada ratusan jenis keju yang diproduksi di
seluruh dunia.[1] Keju memiliki gaya dan rasa yang berbeda-beda, tergantung jenis air susu yang
digunakan, jenis bakteri atau jamur yang dipakai dalam fermentasi, lama proses fermentasi maupun
penyimpanan ("pematangan").[1] Faktor lain misalnya jenis makanan yang dikonsumsi oleh mamalia
penghasil susu dan proses pemanasan susu.[1] Ada lima tahapan utama dalam pembuatan keju.[2]
Peranan DNA Rekombinan Dalam Pembuatan Keju
Pada umumnya keju disukai banyak orang. Keju dibuat dari air susu yang diasamkan dengan
memasukkan bakteri, yaitu Lactobacillus bulgarius dan Streptococcus thermophillus. Untuk
mengubah gula susu (laktosa) menjadi asam susu (asam laktat) susu dipanaskan terlebih dahulu pada
suhu tertentu dengan maksud untuk membunuh bakteri yang berbahaya agar berhasil dalam proses
pembuatannya. Selanjutnya, ditambahkan campuran enzim yang mengandung renin untuk
menggumpalkan susu sehingga terbentuk lapisan, yaitu berupa cairan susu yang harus dibuang,
sedangkan bagian yang padat diperas dan dipadatkan. Enzim tersebut akan menambah aroma dan
rasa, juga akan mencerna protein dan lemak menjadi asam amino.
Hal ini sudah terjelaskan pada proses pembuatan diatas. Yang akan dijelaskan kali ini adalah
bagaimana peranan DNA rekombinan dalam Pembuatan keju.
Industri keju memerlukan rennet, yaitu enzim protease yang diperoleh dari lambung anak sapi yang
masih menyusu dan belum makan rumput. Enzim ini digunakan dalam penggumpalan susu. Pada
tahun 1960 FAO memprediksi akan adanya kekurangan rennet karena kebutuhan daging dunia
mendorong peternak untuk memelihara sapinya hingga dewasa. Usaha untuk mencari enzim protease
pengganti rennet dimulai.

Keasaman berpengaruh terhadap kestabilan kasein baik secara langsung maupun tidak langsung

Beberapa mikroba yang dapat menghasilkan enzim sejenis rennet adalah : Mucor meichei, M.
pusillus, Edothia parasitica dan Bracillus subtilis.
Chymosin (rennet hasil rekayasa genetika)
Chymosin (rennet hasil rekayasa genetika) dihasilkan pada awal tahun 1980an dari mikroba
(Escherichia coli, Kluyveromyces lactis dan Aspergillus niger) yang direkayasa. Chymosin telah
mengalami pengujian yang ketat untuk menjamin keamanan penggunaannya. Chymosin memiliki
sifat dan fungsi yang sama dengan rennet yang diperoleh dari anak sapi. Enzim ini dapat digunakan
untuk menghasilkan keju yang kualitasnya sama dengan keju yang dihasilkan menggunakan rennet
dari anak sapi yang lebih baik daripada jika menggunakan rennet dari jamur atau hewan selain sapi.
Chymosin pertama kali digunakan untuk pembuatan keju pada tahun 1988. Pada saat ini hampir 90%
rennet dihasilkan dari khamir terekayasa. Penggunaan Chymosin didukung oleh kelompok
vegetarian dan kelompok agama yang menolak konsumsi bagian tubuh sapi.

Whey tersusun atas laktosa, persenyawaan nitrogen (protein, peptida dan asam amino), abu dan
lemak. Komponen utama yang terdapat dalam protein whey adalah -lactoglobulin (-lg) dan lactalbumin (-la), karena kedua protein ini mempunyai proporsi berkisar 80% dari seluruh protein
whey. Protein whey adalah protein yang alami dan berkualitas tinggi berasal dari susu. Protein ini
terdiri atas asam-asam amino yang dibutuhkan oleh tubuh untuk sintesis protein otot. Fermentasi
laktosa pada whey akan menghasilkan asam laktat, etanol, asetaldehid, diasetil, aseton, asetoin, asam
asetat, dan asam organik lain. Senyawa hasil fermentasi tersebut tentunya akan berpengaruh terhadap
kadar laktosa, keasaman dan jumlah bakteri asam laktat. Semakin banyak asam laktat yang
dihasilkan maka keasaman akan naik dan pH akan turun. Penelitian tentang fermentasi pada whey
sudah mulai dilakukan, diantaranya penelitian tentang whey yang difermentasi dengan Lactobacillus
acidophilus dan L. bulgaricus (Skudra et al., 1998); pertumbuhan dan ketahanan bakteri probiotik
pada whey rekonstitusi (Dgarlic et al., 2005); perubahan yang terjadi pada karbohidrat dan protein
pada whey yang difermentasi oleh bakteri termofilik (Pescuma et al., 2008). Proses fermentasi
menggunakan bakteri B. bifidum yang merupakan bakteri probiotik yang bermanfaat bagi kesehatan.
Pertumbuhan bakteri tersebut sangat dipengaruhi kandungan nutrisi yang terdapat di dalam bahan.
Bakteri tersebut akan menggunakan nutrisi yang terkandung dalam bahan untuk mengubahnya
menjadi asam. Asam yang terbentuk merupakan hasil fermentasi bakteri asam laktat dengan
mengubah laktosa menjadi asam laktat. Menurut Chandan et al. (2006), Bifidobacterium merupakan
organisme saccharolitik yang mampu menghasilkan asam asetat dan asam laktat pada kondisi
anaerob. pH optimum untuk pertumbuhan bakteri ini adalah 6.0-7.0. Suhu inkubasi optimum bakteri
B. bifidum adalah 37-41C. Faktor lama inkubasi dapat mempengaruhi proses fermentasi yang
terjadi, karena dapat mempengaruhi pembentukan asam laktat yang merupakan produk dari proses
fermentasi (Mortazavian et al, 2006). Pertumbuhan bakteri probiotik B.bifidum pada whey
rekonstitusi mengindikasikan bahwa terdapat pengaruh nyata antara faktor lama inkubasi 6, 12, 18,
dan 24 jam pada suhu inkubasi 37C terhadap total bakteri asam laktat yang dihasilkan (Drgalic et
al., 2005). Lama inkubasi 6 24 jam pada penelitian ini diharapkan B. bifidum dapat tumbuh dengan
baik, sehingga semakin lama waktu inkubasi diduga total BAL semakin meningkat, diikuti produksi
asam laktat yang semakin tinggi dan penurunan kadar laktosa.
Biakan Biang
Biakan biang merupakan faktor penting dalam pembuatan keju. Biakan ini memiliki beberapa peran.
Dua tipe utama biakan yang digunakan dalam pembuatan keju:
Biakan yang paling sering digunakan adalah biakan turunan campuran (mixed-strain), dimana dua
atau lebih turunan bakteri mesophilic dan thermophilic berada dalam simbiosis mutualisme yang
saling menguntungkan. Biakan ini tidak hanya memproduksi asam laktat tetapi juga komponen
aroma dan CO2. Karbondioksida sangat penting untuk menciptakan rongga-rongga di tipe keju
butiran dan tipe mata bundar (round-eyed) . Contohnya keju Gouda, Manchego dan Tilsiter dari
biakan mesophilic dan Emmenthal dan Gruyre dari biakan thermophilic .
Biakan turunan tunggal (single-strain) terutama digunakan ketika obyek dipakai untuk
mengembangkan asam dan berkontribusi terhadap degradasi protein, misalnya pada keju Cheddar
dan tipe keju yang sejenis.
Tiga sifat biakan biang yang paling penting dalam pembuatan keju yaitu:
Disamping asam propionat dan asam asetat, terbentuk karbondioksida dengan jumlah yang
signifikan, dimana merupakan penyebab langsung pembentukan mata bundar yang besar pada tipe
keju yang disebutkan di atas.
Laktat juga bisa dipecah oleh bakteri asam butirat, jika kondisinya sebaliknya tidak bagus untuk
fermentasi ini, dimana terbentuk hidrogen sebagai tambahan asam lemak dan karbondioksida yang
volatil tertentu. Fermentasi ini timbul pada tahap akhir, dan hidrogen dapat menyebabkan keju
menjadi rusak.
Fermentasi laktosa disebabkan oleh adanya enzim laktase dalam bakteri asam laktat.
Dekomposisi protein
Pematangan keju, terutama keju keras, dicirikan pertama dan terutama oleh dekomposisi protein.
Level dekomposisi protein mempengaruhi kualitas keju sampai tingkat yang signifikan, kebanyakan
mengenai konsistensi dan rasa. Dekomposisi protein dihasilkan oleh sistem enzim dari
rennet

Satu-satunya efek rennet adalah untuk memecah molekul parakasein menjadi polipeptida.
Pemecahan pertama oleh rennet membuat kemungkinan dekomposisi kasein yang lebih cepat melalui
aksi enzim-enzim bakteri daripada jika enzym-enzym ini harus memecah molekul kasein secara
langsung. Pada keju dengan suhu masak yang tinggi, keju yang dibakar seperti Emmenthal dan
Parmesan, aktifitas plasmin memainkan peranan pada pemecahan pertama.
Pada keju-keju yang halus-sedang seperti Tilsiter dan Limburger, dua proses pematangan saling
terjadi secara paralel, yaitu proses pemasakan normal pada rennet keju keras dan proses pemasakan
pada hapusan (bakteri) yang terbentuk di permukaan. Pada proses yang disebutkan terakhir,
dekomposisi protein berproses lebih jauh sampai akhirnya ammonia diproduksi sebagai hasil aksi
proteolitik yang kuat dari hapusan bakteri.
Metabolisme Produksi Asam Laktat
Secara umum bakteri asam laktat dapat dibedakan atas 2 kelompok yaitu bakteri homofermentatif
yang meliputi proses glukosa difermentasi menghasilkan asam laktat sebagai satu-satunya produk.
Contoh :Streptococus, Pediococcus, dan beberapa Lactobacillus. Bakteri heterofermentatif yaitu
glukosa difermentasikan selain menghasilkan asam laktat juga memproduksi senyawa-senyawa
lainnya yaitu etanol, asam asetat. Contoh : Leuconostoc.
Bakteri homofermentatif melibatkan aldolase dan heksosa aldolase namun tidak memiliki
fosfoketolase serta hanya sedikit atau bahkan sama sekali tidak menghasilkan CO 2. Jalur
metabolisme dari yang digunakan pada homofermentatif adalah lintasan Embden-Meyerhof-Parnas
untuk menghasilakn piruvat kemudian direduksi menjadi asam laktat melibatkan enzim laktase
