experiments, Cell culturing, Microencapsulation, expression of site-specific engineered genes and
purification of recombinant and native proteins etc.
Area of research:
“Molecular Characterization of Reshuffled Bile Salt Hydrolase (Bsh) and Effect of Dietary
Inclusion of Bsh+ and Bsh- Indigenous Probiotic Lactobacillus plantarum strains of Human
Origin on Cholesterol Metabolism of Rats” under the supervision of Dr. Hemalatha R. (Head
& Deputy Director).
2. Organisation: Proteomics and Structural Genomics Lab, NDRI, Karnal
Duration: August 07, 2009 – April 30, 2010
Designation: RESEARCH ASSOCIATE, worked on isolation and assessment of the antibacterial and antifungal
activity of the novel bioactive peptides generated from LAB, cloning & expression of peptidases,
immobilization and purification of proteins like aminopeptidase, carboxylpeptidase, anti-microbial
peptides etc.
Area of research:
“Novel Approaches for Production of Nutraceuticals from Milk and Indian Herbs for
Potential Use in Functional Dairy Foods” under the supervision of Dr. J.K. Kaushik (Co-PI)
3. Organisation: Molecular Biology Unit, D.M Division, NDRI, Karnal
Duration: August 2005 – July 2009
Designation: Ph.D SCHOLAR, worked on screening of lactobacilli, Development of Bsh assays, cloning,
sequence analysis, expression and purification of recombinant and native Bsh protein. Carried out in-
vivo & in-vitro trials for anti-hypercholesterolemic effect of Bsh+ probiotic Lactobacillus in SD rats.
Area of research:
“Molecular cloning, sequence analysis and expression of bile salt hydrolase (Bsh) from a
potential indigenous probiotic Lactobacillus of human origin” under the supervision of Dr.
V.K. Batish (Former Head & Er. Scientist).
TECHNICAL SKILLS
• Purification and characterization of recombinant and native proteins, bioactive proteins with Gel filtration,
Affinity, Ion-exchange, Hydrophobic interaction chromatography on FPLC system (Pharmacia & Akta
Purifier 10), Ultrafiltration, Rotophor (Isoelectric focussing), denaturing and nondenaturing PAGE.
Localization and identification of proteins by Western blotting and Zymograms.
• Cloning, characterization, sequence analysis of gene sequences and optimization of recombinant protein
expression. PCRs (Bsh/Species/Genus/Gradient/RAPD), agarose gel electrophoresis.
• Antimicrobial agar well assay, microtitre plate assay, Spot on lawn assay, ELISA or Microtitre plate reader
• Nucleotide and amino acid sequence submission to NCBI. Efficient use of BLAST, FASTA, ClustalW
implemented in Geneious Pro (4.6.4) for sequence analysis, characterization and Primer designing. Nucleic
acids isolation and purification. Optimization of different Colorimetric Assays. Anaerobes (Bacterial)
culturing in MK3 Anaerobic Workstation (Don Whitley).
• Maintenance and Experimental handling (Survival bleeding, faecal analysis, Dissection etc) of Spargue-
Dawley rats (Male) for hypocholesterolemic studies. Plasma HDL-, LDL-, VLDL-, Total Cholesterol and
TG estimation. Interpretation of data by statistical analysis.
• Bacterial microencapsulation/immobilization, maintenance, enumeration, preservation and propagation of
bacteria from diverse nature of samples. Characterization of Lactobacillus strains for various in-vitro
probiotic attributes, efficient handling of all kind of microbiological work.
RESPONSIBILITIES
• Designing new projects and Preparation of ICB (International Competitive Bidding) bid documents and
technical specifications for purchasing instruments like Fermentor, Anaerobic Chamber, Vacuum
Concentrator, Multi-plate reader, Refrigerated centrifuges, Microfluidizer, Water Purification System, Gel-
Documentation System, Incubator Shaker, HPLC, Inverted Fluorescent Microscope etc. for International
Competitive Bidding (ICB). Demonstration of laboratory to national and foreign visitors. Maintenance of
laboratory and its instruments.
Computer skills
Operating systems: Windows 7, Windows Vista, XP, 2000 and 98.
Software tools: Geneious Pro (4.6.4), DNA Star, Gencheck, Sequin, SYSTAT etc.
