Abstract:
The involvement of genetic factors in Diabetes Mellitus Type 2DMT2 has explained the
importance of genetic studies. Mutations in the hypervariable 1 of the D-loop region mtDNA
might have an important effect in the copy numbers and gene expressions of the
mitochondrial genome since d-loop region is known has regulatory role on mtDNA
replication and transcription. It could disturb mitochondrial functions such as oxidative
phosphorylation and ATP production. The aim of this research was to analyze the mutation of
hypervariable region 1D-loop mtDNA of DMT2 patients. This research screened the d-loop
mitochondrial region in 16 people consisted of 4 diabetic patients that indicated by new kind
of mutations in the gene encoding tRNA mtDNA and one descendant of each patient, and 4
diabetic patients without new mutations in the gene encoding tRNA mtDNA as well as the
descendant of each patient. A total of 30 mutation regions was discovered within the 442 bp
(16.02416.365). The most frequent regions were i.e., T16296C (66.6%), C16126T (61.1%),
C16223T (55.5%), T16294C (55.5%). Sequencing alignment result between patient diabetes
melitus type 2 which has new mutations in the gene encoding tRNA mtDNA and patient
without mutation shows that the average of similarity was 98.4%. Moreover, Sequencing
alignment result between patient diabetes melitus type 2 with their descendent shows that the
average of similarity was 98.8%.
