Are There Really Bacteria Living In My

Yogurt
Lab report

Introduction :
Micro-biome is the microorganism that live in environment or inside our body. Microbiome typically lives in alimentary canal, mostly in the large intestine where is an
anaerobic environment or lack of air. Our body can get the micro-biome from food that
we eat for example, yogurt, kimchi, miso soup, fermanented soy products or probiotics
pill. Also, we can get from the environment around us, from mother when you were born

and mothers milk. Theres also factors that can distrup the growth of bacteria for
instance, when we take antibiotic or NSAIDS, lack of sleep, stress, smoking, dring
alcohol or do drugs. The Important functions of micro-biome are helping for supporting
immune system, protecting from disease and detoxify our body. Bacteria such as
lactobacillus can help us break down food, absorb nutrients, and fight off organisms that
might cause diseases such as diarrhea. Lactobacilus bulgaricus were the most common
bacteria found in yogurt, also Streptococcus thermophilus, too.The significant
preparation process for this experiment are first, we have to sterilize all the materials
that we are using. Also, we need to make sure that the atmosphere is anaerobic which
mean that there are no oxygen to provide the growth of bacteria.

Purpose :
Trying to grow bacteria in our agar plates from different type of yogurt and use water as
a control.

Hypothesis :
If we grow bacteria in water, there will be no bacteria grow up because we use water as
a control. If we grow bacteria in activia and betgen, there will be bacteria grow up.

Material :
- Lab coats
- Gloves
- Clay
- Plastic container with lid
- Water
- Glass spreader
- Marker
- Alcohol

- Goggles
- Agar plates
- Candles
- Yogurt : Actevia, Betagen, Bio
- Medicine dropper
- Paper towel
- Lamp

Method/Procedure :
1. Put on lab coat, gloves, and goggles
2. Obtain medicine dropper, glass spreader, 3 agar plates
3. Moisten a paper towel with 70% isopropyl rubbing alcohol to prevent bacterial contamination
4. Place your 3 agar plates on the paper towel. Do not open them.
5. Using a marker, label the bottom of each agar plate 1-3
6. Suck up rubbing alcohol into the medicine dropper. Hold for 10 seconds. Squirt it out onto a
paper towel. Use the paper towel to rub the outside of the dropper.
7. Hold the dropper over the lamp to burn off the alcohol
8. Set the dropper down but propped up onto a paper towel so that the tip of the dropper does
not touch anything.
9. Obtain the diluted yogurt (yogurt mixed with water)
10. Using the sterilized dropper suck up yogurt to fill the dropper. Do not touch anything other
than the yogurt. Continue to hold the dropper in one hand
11. Open 2 agar plates and add a few drops of the yogurt to the plates, one at a time. Add the
drops into the center of the plates, one drop on top of the other.
12. Do not set the lids to the agar plates down on anything
13. Put the lids back onto the agar plates

14. Onto the 3rd agar plate, add a few drops of distilled water, for the control. (either with new
dropper or teacher will provide drops for you) 15. Dip the glass spreader into rubbing alcohol for
10 seconds. Hold it over the lamp to burn off the alcohol.
16. Do not let the triangular part of the glass spreader touch anything. Hold it in your hand the
whole time.
17. Open each agar plate (do not put the lids down) and use the glass spreader to spread the
diluted yogurt in a specific pattern. Start with agar plate 3 to prevent contamination on the
control with diluted yogurt. Do not push too hard so that you break the agar. 18. After you finish
each plate, quickly put its lid back on. Wipe the glass spreader clean between each plate.
19. Let the plates sit for a minute to help the yogurt attach to the agar.
20. The lab instructor will prepare an anaerobic chamber to place all of the groups agar plates
into. The anaerobic chamber consists of a plastic container and a candle. When the candle is lit,
place the lid onto the container and cover the outside with clay. The candle will eventually go
out, leaving a container with no oxygen.
21. Wait a few days, then check on your agar plates

Result:
We were successful in growing bacteria in the Actevia. There was a white colony of bacteria
including bacteria that we wanted to see and didnt , but there was mold because of the aerobic
environment after the experiment. Unfortunately, Our water was contaminated. We thought that
our materials might not were well sterilized and we might open the lid for too wide which allowed
bacteria in the air went inside. Becuase of water was our control and the bacteria was grew in
water, that ruined the experiment. The reason is water was not suppose to have bacteria. So,
the 2 samples of yogurt might be contaiminated and had bacteria from the environment mixed
with the bacteria in yogurt,too. We must well sterilize and be careful when we do the experiment
next time. For our experiment in growing bacteria in Betagen, we had observed and found out
that the bacteria was a long white colony all over the agar plate.

Conclusion:
Bacteria or microbiome that lives in our digestive system can get from our mother when we
were born, mothers milk or food that we ate like fermanented food. Micriobiome has many
important roles in our digestive system for example, it helps to digest and absorb nutrients. The
purpose of this experiment is to grow bacteria that lives in 2 brands of yogurt including Betagen
and Actevia. Also, we used water as a control. The most important procedure of this experiment
was to neutralize all the tools that we used, so that it didnt have any bacteria alive. We were
successful in growing bacteria in the Actevia. There was a white colony of bacteria, but there
was mold because of the aerobic environment after the experiment, but there was still other
bacteria that grew up,too and that made us unsure that it was bacteria from yogurt or from the
environment. Unfortunately, Our water was contaminated. We thought that our materials might
not well sterilized, we might not dry off all alcohol and we might open the lid for too wide which
allowed bacteria in the air went inside. Because of water was our control, but the bacteria was
grew in water and that ruined the experiment. The reason is water was not suppose to have
bacteria in it. So, the 2 samples of yogurt might be contaiminated and had bacteria from the
environment mixed with the bacteria in yogurt,too. This experiment made us learned that we
have to follow the procedures carefully and make sure that everything well sterilized, also we
had to be neat and tidy, so that no bacteria went into yogurt. Moreover, we had to use fire to
sterilize and that was very dangerouse and we learned that we must listen to the lab technician
and follow the instructions with full attention.
