Materials and methods

For the seed germination and seedling growth experiment, 15 pairs of petri dishes
lined with filter paper were used as container of the germinating seed. Several
reagents were prepared such salt solutions of different concentration, gibberellic
acid solution, benzyl amino purine, and naphthalene acetic acid and were used to
observed their effect in germination and growth of the mung bean seed.

A. Effect of Temperature

For the observation of the effect of temperature, three pairs of cleaned ptetri
dishes were prepared using filter paper as lining and were moisten with 10 ml of
distilled water. 10 equally sized mung bean seeds were place in each of the petri
dishes. The petri were set in three different places; at room temperature, at the
refrigerator and at the oven. The temperature in those places were recorded. After
two days the percent germination was recorded and the measurement of the
hypocotyl-root axis was measured.

B. Effect of pH

For the effect of pH same set up were used except for the other two petri dishes 10
ml of pH 3 and pH 11 treatment solution were used instead of distilled water to
moisten the filter paper. The pH of the three solution were measured using pH
paper. 10 uniformly sized mung beans seeds are place in each petri dishes. The
hypocotyl-root axis wear measured and the percent germination was recorded after
two days.

C. Effect of Varying Osmotic Pressure

For the effect of varying osmotic pressure, same set up with A were prepared except
that other two petri dishes were moisten with 10 ml of 1% and 1.5% of NaCl
solution. 10 uniformly-sized mung bean seed were place in each o the three dishes.
After two days, the hypocotyl-root axis was measured and the percent germination
was calculated.

D.Effect of light

For the effect of light, two pairs of cleaned petri dishes were prepared. 10 mung
bean bean seeds were placed in each dish. One of the petri dishes were set in the
place were there is light and the other one were put inside the locker. The
hypocotyl-root axis was measured and the percent germination was calculated after
six days.

E. Effect of Hormones

Lastly for the effect of hormones, four pairs of cleaned petri lined with filter paper
were prepared. 10 ml of distilled water, gibberellic acid solution, naphthalene acetic
acid, and benzyl amino purine solution were used to moisten the dishes. 10 equally-
sized mung bean seeds were place in each of the dishes. The dishes were set in
dark area. After two days, the hypocotyl-root were measured and the percent
germination was calculated.
