1. Safety
1.1 Instrument Disinfection
It is very important that the instrument has been thoroughly disinfected before
performing the tests provided in this IFU.

WARNING
THE DISINFECTION PROCEDURE SHOULD BE PERFORMED
ACCORDING TO NATIONAL, REGIONAL, AND LOCAL REGULATIONS.

WARNING
ALL PARTS OF THE INSTRUMENT THAT COME INTO CONTACT
WITH POTENTIALLY INFECTIOUS MATERIAL MUST BE TREATED
AS POTENTIALLY INFECTIOUS AREAS.

IT IS ADVISABLE TO ADHERE TO APPLICABLE SAFETY


PRECAUTIONS, (INCLUDING THE WEARING OF POWDER-FREE
GLOVES, SAFETY GLASSES AND PROTECTIVE CLOTHING) TO
AVOID POTENTIAL INFECTIOUS DISEASE CONTAMINATION WHEN
PERFORMING THE DISINFECTION PROCEDURE.

1.1.1 Disinfection Procedure


The instrument should be disinfected using a solution of 70% ethanol.

RISK OF FIRE AND EXPLOSION!
PRIOR TO CLEANING THE OUTER SURFACE OF THE INSTRUMENT
AND THE DISPLAY, SWITCH OFF THE INSTRUMENT AND DISCONNECT
IT FROM THE MAIN POWER SUPPLY!
ETHANOL IS FLAMMABLE AND WHEN IMPROPERLY HANDLED CAN
LEAD TO EXPLOSIONS AND/OR FIRE. PROPER LABORATORY SAFETY
PRECAUTIONS MUST BE OBSERVED.

Caution
Before starting the disinfection procedure
use a rinse procedure (Rinse Day)
with distilled or deionized water (lab quality) to flush the system.

WARNING
THE DISINFECTION PROCEDURE SHOULD BE PERFORMED IN A
WELL-VENTILATED ROOM BY AUTHORIZED TRAINED PERSONNEL
WEARING DISPOSABLE POWDER-FREE GLOVES, PROTECTIVE
GLASSES AND PROTECTIVE CLOTHING.

Please note that the disinfectant can influence the performance of your
instrument if it comes into contact with the electronics!
The following procedure should be used to disinfect the instrument:
1. Wear protective powder-free gloves, protective glasses and protective
clothing.
2. Prepare an autoclave bag for all disposables used during the disinfection
procedure, label it with autoclave tape and put it in the autoclave or follow
the procedure defined in chapter 1.1.2 Disposal of Operating Material.
3. Prime the liquid system with disinfectant. Perform the Rinse - Night
procedure with disinfectant solution and stop the procedure after five
hours.
4. Switch off the instrument and disconnect the instrument from the main
power supply.
5. Disconnect the instrument from any accessories that are used for
example: Liquid Level Detection System, computer, etc. Accessories that
should be shipped together with the instrument have to be included in the
disinfection procedure.
6. Carefully spray the disinfectant solution (or use a disposable soft tissue
paper towel soaked in the disinfectant) on all outer surfaces of the
instrument (continued on next page).
7. After a minimum contact time of 10 minutes, repeat step 6 of this
procedure.
8. Wipe dry the outer surfaces of the instrument.
9. Wash your hands with a mild detergent and then disinfect them.

1.1.2 Disposal of Operating Material

WARNING
CHEMICAL AND BIOLOGICAL HAZARDS CAN BE ASSOCIATED WITH
THE WASTE MATERIAL (MICROPLATE) OF PROCESSES RUN ON THE
HYDROFLEX.
TREAT THE USED MICROPLATE, WASTE BOTTLE, PRIME TRAY ON
THE PLATE CARRIER, DISPOSABLES AND ALL SUBSTANCES USED,
IN ACCORDANCE WITH GOOD LABORATORY PRACTICE GUIDELINES.
INQUIRE ABOUT APPROPRIATE COLLECTING POINTS AND
APPROVED METHODS OF DISPOSAL IN YOUR COUNTRY, STATE OR
REGION.

2.1 Specifications

2.1.1 Hardware Requirements


The following hardware requirements have to be met to use the XFlex service
software:

2.2 Installation

2.2.1 Installing the Program

Caution
STOP It is very important that the person who installs the XFlex service
software has System Administrator rights on the computer.

XFlex is installed by a software setup program. All of the necessary components


are installed automatically.
The software is installed using the following procedure:
1. Start the computer.
2. Insert the XFlex service Software CD into the necessary CD drive or
download the XFlexService.exe file from the Tecan ftp-server (contact
your local service representative for details). Once the file has been
downloaded continue from step 5.
3. When using a CD, an Explorer window will automatically open. The CD is
named TECAN and TECAN will appear as the title of the Explorer
window.
4. The setup program is located using the following path:
D:\Software\XFlex_Vx_x\ XFlexService.exe, where D: is the CD drive
that contains the XFlex service Software CD and x_x is the version
number of the XFlex service software.
5. Double-click the XFlexService.exe file and the installation program is
started.
6. A series of dialog boxes will appear, read each one, enter any necessary
information and click Next to continue.
7. The files are then installed and the program icon is created.
8. When the Installation Complete dialog box appears, click Finish and the
program is ready to be used.
9. When using a CD, store the XFlex service Software CD in a safe place in
case the software needs to be reinstalled.

2.2.2 Uninstalling the Program

CAUTION
STOP IT IS VERY IMPORTANT THAT THE PERSON WHO UNINSTALLS THE
XFLEX SOFTWARE HAS SYSTEM ADMINISTRATOR RIGHTS ON THE
COMPUTER.

The uninstalling procedure is offered automatically when starting an installation of


a new software version. It can also be started from the Windows Start menu:
Start/Settings/Control Panel/Add or Remove Programs. Find the XFlex software
in the list of currently installed programs and click Remove.

1. The following message will appear asking you if you really want to
remove the program. Click Yes to continue.

2. The following message will appear asking if you would like to remove all
created configuration and instrument data files. Click Yes or No.

3. The program is uninstalled and is no longer listed in the Add or Remove


Programs window.

2.3 Starting XFlex


Start the program from the Start menu: Programs/Tecan/XFlex.
The Connect command from the Instrument menu can be selected only when
the Program menu is visible on the instruments display.
XFlex service software can also be used in simulation mode.

It is possible to use XFlex with instruments with firmware versions lower than 2.0;
however this is not recommended, as some of the tests will not be able to be
performed. It is recommended to update the firmware version of the instrument to
Version 2.0 or higher.

2.4 Connecting the HydroFlex Washer


After installing and starting the HydroFlex software, connect the instrument to the
computer and switch ON the instrument.
The Found New Hardware notice should appear.

Click on this notice and the following dialog box appears:

Select No, not this time and click Next to continue.

The following dialog box appears:

Select Install from a list or specific location (Advanced) and click Next to
continue. The following dialog box appears:

Select Dont search. I will choose the driver to install. and click Next to
continue.

Select Show compatible hardware and click Next to continue. The necessary
configuration tasks are performed. Once all the tasks have been performed
successfully, the following dialog box appears:

Click Finish to close the wizard.


The XFlex service software can now be used.

3.1.3 Toolbar Description


The program has the following buttons:

Click the Connect button to connect to the instrument.

Click the Disconnect button to disconnect to the


instrument.

3.1.4 Tabs/Control Pane and Info Pane Description


The tabs provide the same function as the menus with the same names. Each tab
opens a page in which a list of tests appears. To the right of each test name are
three items: the Status field, the Details button and the Run button.
The Status field displays TODO, WORKING, DONE, PASSED or FAILED
depending on the status or result of the test.
Click the Details button to open the Info pane in which details about the current
status and results of the test can be viewed. Click the Close button to close this
pane in order to view other details or perform tests.
Click the Run button to open the corresponding perform the test.

3.1.5 Status Bar Description


The status bar provides the Instrument Connection Status, Instrument Serial
Number and the Instrument Name.

3.2 Log Entry


The Log Entry dialog box appears before each test or adjustment procedure is
performed.

Enter the User name and the reason for performing the test or adjustment.
If a part has been replaced, select Test after replacing part and enter the name
of the part in the field.
If no parts were replaced, select Other and enter the reason for performing the
test or adjustment in the field.

3.3 Exiting XFlex


To exit the XFlex software, select Quit from the File menu or click the close
button in the upper right hand corner of the main window.

4.1 File Menu

4.1.1 Change User


Select Change User from the File menu to logout the current user and login a
new user. The following dialog box appears:

Enter the User name and click OK.

4.1.2 Options
Select Options from the File menu to open the Options dialog box:

The paths of the Configuration File, DHR Main Directory, Firmware Download
and Language Download can be viewed.

The paths can also be modified by clicking the Browse button and browsing
for the desired folder.
Click OK to save changes or Cancel to discard changes.

Select Quit to exit the XFlex software.

4.2.1 Disconnect/Connect
Select Connect/Disconnect from the Instrument menu to connect or disconnect
the instrument.

4.2.2 Prime
Select Prime from the Instrument menu to prime the instrument.

Select the Channel (1 -4), duration Time (5 99 s) or Volume (15 820 ml) and
click Start to begin the priming procedure.
Click Stop to abort the procedure. When the prime procedure has ended, the
Prime dialog box closes automatically.

4.2.3 Rinse
Select Rinse from the Instrument menu to perform a rinsing procedure.

Select the Mode (Day or Night), the Channel (1 -4) or duration Time (5 99 s)
or Volume (15 820 ml) and click Start to begin the rinsing procedure.
Click Stop to abort the procedure. When the rinse procedure has ended,
click Close to return to the main window.

4.2.4 Send Command


Firmware commands can be sent to the instrument and responses will be
displayed in the received data window.

For example, enter ?VERSION in the Command String field and click Send to
send the command.
The response appears in the Received Data window, e.g.

4.3 Store/Restore Menu

4.3.1 Instrument Settings


Select Instrument Settings from the Store/Restore menu in order to read the
current instrument parameter values and write them to DHR (see 3.1.2 Options
for information about setting the default path for DHR). Previously saved
instrument parameter values can also be read from DHR and written to the
instrument.

For Firmware Versions 2.01 to 2.05:

Read Instrument Reads current instrument parameter values from the


button instrument
Write to DHR Writes currently read instrument parameter values to
button DHR
Read from DHR Reads previously stored instrument parameter values
button from DHR
Set Instrument Writes instrument parameter values from DHR to the
button instrument
Close button Closes the dialog box

For Firmware Versions Higher Than 2.05:

Read Instrument Reads current instrument parameter values from the


button instrument
Write to DHR Writes currently read instrument parameter values to

Read from DHR Reads previously stored instrument parameter values

Set Instrument Writes instrument parameter values from DHR to the
button instrument
Close button Closes the dialog box

Select SavePlatesAndPrograms from the Store/Restore menu in order to
upload the plate and program parameters currently stored on the instrument to
DHR (see 3.1.2 Options for information about setting the default path for DHR).

instrument.

4.4 Setup Menu

4.4.1 Serial Number


1. Select Serial Number from the Setup menu to set the Material Number,
Serial Number and Instrument Alias.

2. Select the Instrument Type from the drop-down list. The Serial Number
has to be entered manually. The Material Number and Instrument Alias
fields will be filled in automatically. If your instrument is not listed, select
Other and fill in the fields manually.
3. Click Set to confirm changes and the following dialog box appears:

4. Click OK, switch the instrument OFF and ON again and reconnect the
instrument to the software.

4.4.2 Firmware Download

1. Select Firmware Download from the Setup menu to download new


firmware to the instrument.

2. Select the source path by clicking the browse button .

3. Select the file type from the list under Download: Main, Display,
Process Control, or Vacuum Filtration.

4. Click Download to download the selected firmware to the instrument.

5. When downloading the Main firmware, the following dialog box appears:

6. Unplug the USB cable and plug it in again and click OK.

4.4.3 Language Download


1. Select Language Download from the Setup menu to download
language files to the instrument.

2. Browse for the source file by clicking the browse button .


3. Click Download Language File to download the selected language files

5. If the language file has been successfully downloaded, DONE will appear
in the status field next to Language Download.

4.4.4 Instrument Configuration


1. Select Instrument Configuration from the Setup menu to adjust the
instrument configuration according to the instrument connected.

4.5.2 Process Control Test


1. Select Process Control Test from the Options menu to check the
function of the process control.

2. Connect channel 1 to a container containing wash buffer and click the


upper Prime button to prime channel 1 with wash buffer.
3. Click Start wash buffer test.
4. When the test is complete, PASSED or FAILED appears.
5. Connect channel 1 to a container containing distilled water and click the
lower Prime button Prime channel 1 with distilled water.
6. Click Start distilled water test.
7. When the test is complete, PASSED or FAILED appears.
8. The following dialog box appears after completing the test:

9. The Process Control module is deactivated after the test. If necessary,


reactivate the Process Control module from the Instrument
Configuration menu.
10. Click OK to close the Information dialog box.

4.5.3 LLD Test


1. Select LLD Test from the Options menu to test the function of the liquid
level detection.
2. Click Start LLD Sensor Test to start the test.

Active sensors will show the status Empty or Full as in the picture below

4.5.6 High VAC Test


1. Select High VAC Test from the Options menu.

Preparation appears in the dialog box when the waste bottle is under
pressure. A valve will be opened and the pressure will be released.

During the first step, the instrument builds up pressure and must reach
the target pressure within 2 minutes.

3. After the target pressure has been reached, place an empty microplate
(Greiner 96-well flat transparent) on the plate carrier.

4. Click OK when ready, to start the second step of the test.


The following dialog box appears:

5. Manually check that the vacuum pressure holds the plate in place on the
carrier. Click Yes or NO to start the third step of the test.
The following dialog box appears:

6. The vacuum pressure will be released. Check that the vacuum is NOT
holding the plate in place anymore. Click Yes or No to start the fourth

4.6 Function Test Menu

4.6.1 Y, Z Motors
1. Select Y, Z Motors from the Function Test menu to check the function of
the Y and Z motors.

2. Move the Y motor counterclockwise and clockwise by clicking the


appropriate buttons. Click PASSED if the motor moves correctly,
otherwise click FAILED.
3. Move the Z motor counterclockwise and clockwise by clicking the
appropriate buttons. Click PASSED if the motor moves correctly,
otherwise click FAILED.
4. Click Start Plate Sensors Test (No vac. plate carriers):
The plate transport will be moved until the plate transport sensor and the
plate sensor (Hall-sensor) are activated. The test is completed
automatically and passes if the motor finds the home positions.
Two green lights will appear when the test has passed:

5. Click Start Manifold Home Position Sensor Test to move the Z motor
to its home position. Click PASSED if the Z motor finds the home
position, otherwise click FAILED.
6. Under Manifold Head Up Sensor, click Start Sensor Test. Activate and
deactivate the sensor manually by lifting and lowering the manifold arm.
Click PASSED if the sensor state is displayed correctly, otherwise click
FAILED.
7. Click Close to close the dialog box.

4.6.2 Dispense Pump


1. Select Dispense Pump from the Function Test menu to check the
function of the dispense pump.

2. Click Start Function Test to start the dispense pump test.


The test has PASSED if the dispense pump runs with a constant rate that
is higher than the specified limit, otherwise the test has FAILED.

4.6.3 Aspiration Pump


1. Select Aspiration Pump from the Function Test menu to check the
function of the aspiration pump.

2. Click Start Aspiration Pump to start the aspiration pump.


3. Click Stop Aspiration Pump to stop the aspiration pump.
The test has PASSED if the aspiration pump starts and stops after
pressing the appropriate buttons, otherwise the test has FAILED.

4.6.4 Channel Valve Test


1. Select Channel Valve Test from the Function Test menu to test the
function of the liquid channels.

2. Click the Start button to start the test for the specified liquid channel.
The instrument will be primed using the selected channel.
3. Check the liquid flow and select PASSED or FAILED for the tested
channel.
4. Click Close after the liquid channels have been tested.

4.6.5 Bubble Sensor Test


1. Select Bubble Sensor Test from the Function Test menu to test the
function of the bubble sensor.

2. Click the Prime button to start the priming procedure (use distilled water).
The following dialog box appears:

3. Select the Channel (1 -4), duration Time (5 99 s) or Volume (15 820


ml) and click Start to begin the priming procedure.
4. Click Stop to abort the procedure. When the prime procedure has ended,
the Prime dialog box closes automatically.

5. Make sure the channel 1 tube is in distilled water, click the Start Bubble
Sensor Test button. The following dialog box appears:

4.6.6 Burn In
1. Select Burn In from the End Test menu to perform the endurance test.

2. Click Enable Burn In. The following dialog box appears:

4. Disconnect the instrument, by clicking the Disconnect button.
5. On the instruments display, perform the following:
PROGRAM Press left or right arrow keys
< > OK until BURN IN TEST appears.

BURN IN TEST Press OK to start the endurance test.

4.7.1 Y Motor Resolution (depending on FW and material


number)
1. Select Y Motor Resolution from the Adjust menu.

3. Insert the adjustment plate (the one with the notches).

manifold until it falls into the notch.

Note
If the manifold is moved too far in the Y or Z direction,
click Initialize Y/Z Motors and repeat the procedure.

10. Click the First Notch Found button to save this as the first position.
11. Move to the last position (notch) by clicking the Move to Last Notch
Default Position button.
12. Move the manifold to the edge of the last notch using the appropriate
Backward XXXX Step(s) buttons.
13. Lower the manifold using the appropriate Down XXXX Step(s) buttons
until the needles rest on the bottom of the notch.
14. Then lower the manifold again by 50 steps using the Down 50 Steps
button.
15. Then use the appropriate Backward XXXX Step(s) buttons to move the
manifold until it falls into the notch.
16. Click the Last Notch Found button to save this as the last position.
17. Save the newly calculated Y motor resolution by clicking the
Set Y Resolution button.

4.7.3 Plate Sensor Offset


1. Select Plate Sensor Offset from the Adjust menu to calibrate and check
the function of the plate sensor.

2. Click Calibrate Plate Sensor to start the procedure. Make sure that the
housing is mounted on the instrument to prevent false calibration results.
3. Insert a microplate and click Check Plate Sensor. The plate should be
detected: a green light and Plate will appear in the dialog box as shown
below:

4. Remove the microplate and click Check Plate Sensor again. The plate
should not be detected: a red light and No Plate will appear in the
dialog box.
5. Click PASSED if the sensor detects the plate correctly, otherwise click
FAILED.

4.7.4 Adjust Prime Position


1. Select Adjust Prime Position from the Adjust menu to check the prime
position and correct it if necessary.

2. Click Move to Current Prime Position to move the manifold to the


currently set prime position.
3. Check the position and, if necessary, correct it using the Movements
buttons.
4. Once the prime position has been properly adjusted, click Save under
Movements to save the new position.
5. Click Test Current Prime Position to make sure the prime position is
correct. Readjust if necessary.
6. If the prime position is correct, click PASSED to confirm and the dialog
box will close.

1. Select Bubble Sensor Calibration from the Adjust menu to calibrate the

4.7.6 Dispense Pump Calibration


1. Select Dispense Pump Calibration from the Adjust menu to calibrate
the dispense pump.

2. Click the Prime button to start the priming procedure (use a solution of
distilled water with 0.1% Tween).
3. Weigh an empty microplate and enter its weight in the appropriate
Plate Weight field (mg). Place the plate on the plate carrier.
4. Dispense 50 l of distilled water into the plate by clicking the appropriate
Start Dispense button.
5. Weigh the microplate and enter its weight in the appropriate
Plate Weight field (mg). Place the plate on the plate carrier.
6. Dispense 250 l of distilled water into the plate by clicking the
appropriate Start Dispense button.
7. Weigh the microplate and enter its weight in the appropriate
Plate Weight field (mg). Place the plate on the plate carrier.
8. Click the Start Aspirate button to aspirate the liquid out of the wells.
9. Click the Calculate Calibration Factors button and the new calibration
factors are calculated and appear in the New Slope and Offset fields.
10. Click Write to Instrument to finish the dispense pump calibration
procedure.
11. The procedure is DONE if no errors occur, otherwise the result is
FAILED.
12. Click Close to close the dialog box.

4.7.7 Dispense Rate Calibration


1. Select Dispense Rate Calibration from the Adjust menu to calibrate the
dispense rate.
The following dialog box appears for firmware versions 2.01 to 2.05:

(The Automatic Run 200 500 l/s button is available in versions > 2.05, except when working in Drip Mode)
2. Click the Prime button to start the priming procedure using channel 1
and a solution of 0.1% Tween.
3. Start each wash procedure by clicking the corresponding Start test: xxx
l/s button.

4. Use a stopwatch to measure the dispense time. Enter the times in the
Time/Result fields.
5. Adjust the voltage value if necessary and click the corresponding Set
button.
6. Repeat the procedure for each speed until dispense time is within the
given range (Expected Time +/- 1 s).
7. For drip mode, check that the liquid is dripping and click OK if drip mode
is adjusted correctly. Dispense time is not measured for drip mode.
8. When the test is complete, click OK or FAILED.
9. The procedure has PASSED if the acceptance criteria have been met,
otherwise the result is FAILED.

4.8 End Test Menu

4.8.1 Verify Motor Positioning


1. Select Verify Motor Positioning from the End Test menu to check the
motor positioning.

2. Adjust the position of the manifold by using the appropriate buttons.


When the manifold is positioned exactly above the crosses on the
adjustment plate.
3. Confirm the positioning by clicking the Manifold in Position button and
the test will be declared PASSED.
4. Click Close to close the dialog box.

4.8.2 Plate Sensor Offset Test


1. Select Plate Sensor Offset Test from the End Test menu to check the
plate sensor offset.

and click OK.
After the first phase of the calibration procedure has finished, the

4.8.3 Rinse Position Test (depending on FW)


1. Select Rinse Position Test from the End Test menu to check the rinse
position.
2. Make sure the manifold is in the working position (down).
3. Start the test by clicking the Start Rinse Position Test button. The
instrument will start a rinse procedure and will fill the prime tub with
liquid.

The test is performed automatically. The test has passed when


PASSED appears in the field next to the Start Rinse Position Test
button.
4. Click Close to close the dialog box after the test has finished. The
instrument will finish the rinse procedure and the prime tub will be
emptied.

4.8.4 Prime Position Test


1. Select Prime Position Test from the End Test menu to check the prime
position.

2. Start the test by clicking the Check Prime Position button.


3. Visually verify that the manifold needles do not touch the inside edges of
the prime tub in the front or back (this applies to all plate carriers).

Needles must not


touch the edges!

4. Click PASSED if the manifold needles do not touch the edges of the
prime tub, otherwise the result is FAILED.
5. Click Close to close the dialog box after the test has finished.

1. Select Verify Dispense Capability from the End Test menu to check the
dispense capability.

2. Click the Prime button to start the priming procedure (use a solution of
distilled water with 0.1% Tween on channel 1).
3. Weigh each of the 8 strips of a Greiner 96-well microplate (12-well
transparent strips) and enter each weight in the appropriate
Weight Strips X field (mg) under Weight Empty Strips.
4. Place the empty microplate on the plate carrier.
5. Dispense 200 l of distilled water/Tween solution into the plate by clicking
the Dispense 200 l button.

6. Weigh each strip and enter each weight in the appropriate


Weight Strip X field (mg) under Weight 12 x 200 l.

7. Place the filled microplate back onto the plate carrier.


8. Click Calculate and the results will appear in the Result group box.

2. Click the Prime button to start the priming procedure (use a solution of
distilled water with 0.1% Tween on channel 1).

4.8.7 Verify Aspiration Capability MBS (depending on material


number)
1. Select Verify Asp. Cap. MBS from the End Test menu to check the
aspiration capability (MBS plate carriers only).

2. Insert the adapter frame into the plate carrier. This is important to prevent
the wells from being affected by the magnets.
3. Click the Prime button to start the priming procedure (use a solution of
distilled water with 0.1% Tween on channel 1).
4. Weigh each strip of a Greiner 96-well microplate (12-well transparent
strips) and enter each weight in the appropriate Strip Weight X field (mg)
under Weight Empty Strips.

5. Place the empty microplate onto the adapter on the plate carrier.

6. Dispense 200 l of distilled water/Tween solution into the plate by clicking


the Dispense 200 l button.

7. Click the Aspirate button to aspirate the liquid out of the wells.

8. Weigh each strip again and enter each weight in the appropriate
Weight Strip X field (mg) under Weight Strips After Aspiration.

9. Click Calculate and the results will appear in the Result group box.

10. The procedure has PASSED if the acceptance criteria have been met,
otherwise the result is FAILED.

11. Click Close to close the dialog box.

Note
The aspiration test may fail if the Y/Z motor offsets have been
