PRICES:
Unless otherwise herein stated, the prices set forth on a purchase order accepted by Seller shall be final and any trans-
portation charges shall be due at the date of shipment. Unless otherwise indicated on the face of a purchase order accept-
ed by Seller, all prices are F.O.B. point of shipment and do not include any cost for freight, which may be prepaid for
Buyer’s account, any costs or charges for insurance or any applicable sales, use, transfer, excise or other taxes, tariffs or cus-
tom duties, all of which are the responsibility of Buyer whether paid by Seller or Buyer.
TERMS OF PAYMENT:
Buyer will be invoiced at the time of shipment of each Product. Buyer shall make payment in full within thirty (30) days of
the date of the invoice. Late payments may incur a charge at the rate of one and one-half percent (1.5%) percent per
month, or the maximum allowed by law, whichever is less. Further shipment of Products may be declined without advance
notice if Buyer fails to make any payment when due, or if the financial condition of Buyer becomes unsatisfactory to Seller.
Seller may elect to retain a security interest in all Products sold to Buyer to secure all of Buyer’s obligations to Seller under
these Terms and Conditions, and Buyer will execute any documents necessary to create and perfect this interest. Sales by
Seller shipped outside the U.S. may require payment on an irrevocable letter of credit reasonably acceptable to Seller.
DELIVERY DATES:
Delivery dates set forth on a purchase order accepted by Seller are subject to change and are predicated on conditions
existing at this time. Seller does not guarantee any delivery dates and shall not be responsible for any loss or damage of
any kind or nature whatsoever caused by any delay in delivery irrespective of the cause of such delay.
FORCE MAJEURE:
In addition to other limitations stated herein, Seller shall not be liable for any delay in Seller’s performance due to fires,
strikes, labor disputes, war, terrorist acts, civil commotion, embargoes, floods, delays in transportation, shortages or failure
of supply, fuel, power of transportation, breakdown of equipment, acts of God, or other circumstances beyond Seller’s rea-
sonable control.
WARRANTY:
Seller warrants that all Products purchased by Buyer will conform to applicable Product specifications at the time of deliv-
ery to Buyer. SELLER MAKES NO OTHER WARRANTY, EXPRESS OR IMPLIED, INCLUDING, WITHOUT LIMITATION, ANY
IMPLIED WARRANTY OF MERCHANTABILITY OR FITNESS FOR A PARTICULAR PURPOSE, ALL SUCH WARRANTIES BEING
HEREBY EXPRESSLY EXCLUDED. IT IS THE SOLE RESPONSIBILITY OF BUYER TO DETERMINE THE ADEQUACY OF ALL PROD-
UCTS PURCHASED BY BUYER FOR ANY INTENDED SPECIFIC PURPOSE OR USE.

LIMITATION OF REMEDIES:
Buyer’s sole remedy, and Seller’s sole liability for non-conforming goods rejected as provided herein, shall be limited to
replacement of the Products or, at Seller’s option, refunding the portion of the price of such non-conforming Products paid
to Seller. In no event shall Seller’s liability for any claim, loss, costs or damages relating to any Products shipped, stored,
Sold or delivered hereunder exceed the purchase price therefore, nor shall Seller be liable for any loss, charges or damages
resulting from its inability to procure any Products ordered by Seller or for delays or failure to deliver Products hereunder.
IN NO EVENT SHALL SELLER BE LIABLE FOR ANY INDIRECT, INCIDENTAL, CONSEQUENTIAL OR SPECIAL DAMAGES,
WHETHER ARISING UNDER CONTRACT, TORT, STRICT LIABILITY, STATUTE OR OTHER FORM OF ACTION, EVEN IF SELLER HAS
BEEN INFORMED OF THE POSSIBILITY THEREOF.
CONFIDENTIALITY:
Buyer will keep in strictest confidence all Confidential Information that is disclosed to Buyer by Seller, which Buyer
acquires, or to which Buyer has access, whether or not such information is marked as “confidential.” In the event of an
actual or threatened breach of this covenant, Seller shall be entitled to injunctive relief restraining Buyer, other persons,
or any combination thereof, from disclosing Confidential Information. Also, Seller may pursue all other available remedies
including, but not limited to, recovery of damages (including reimbursement for attorneys’ fees, accountants’ fees and
expert witness fees), from Buyer. “Confidential Information” includes but is not limited to any information not generally
known to the public (including, without limiting the generality of the following, information about Seller’s operations,
personnel, products or services) which, if misused or disclosed, could have a reasonable possibility of adversely affecting
the business of Seller, whether presently existing or developed in the future: (a) Seller’s business methods, processes, sys-
tems, products, marketing, sales, services, financial information and practices; (b) compilations of data or information con-
cerning Seller’s business and investments; (c) the names and business of Seller’s suppliers and customers and the nature of
Seller’s relationships with such suppliers and/or customers; and (d) confidential, proprietary, or trade secret information
submitted by Seller’s other suppliers, employees, or consultants to Seller for study, evaluation or use.
ENTIRE CONTRACT:
No terms or conditions, other than those stated herein, and no agreement or understanding, oral or written, purporting to
modify these Terms and Conditions, whether contained in Buyer’s purchase order or elsewhere, shall be binding on the
Seller unless made in writing and accepted in writing by Seller.

DETERGENT ANALYSIS
Each lot of Anatrace detergent is analyzed so that you can be assured of the highest consistent quality
available anywhere. Our ANAGRADE® detergents are purified to be greater than 99% pure as measured
by HPLC and to be low in UV absorbing or fluorescent impurities.
We are pleased to list below the analytical procedures used to evaluate our detergents. Should you have any
questions about these procedures, please feel free to contact us.
Some impurities may be less than one percent and still affect the properties of a detergent lot.
Therefore, the following tests are also performed to insure that you receive the highest quality
detergent available.

Absorbance
The absorbance of the detergent solution (1% w/v) in water is measured in the UV region. Glucosides
and maltosides should have low absorbance throughout this region.
Fluorescence
The fluorescence of the detergent solution (0.1% w/v) in water is compared to a standard BSA solution
unless otherwise stated. The excitation wavelength is 280 nm and the emission is measured at 345 nm.
Conductance
The conductance of the detergent solution (10% w/v) in water is routinely measured. For those deter-
gents which are nonionic or zwitterionic, a detergent solution should have conductance nearly the same
as deionized water.
Solubility in water
The solubility of the detergent solution in water is routinely tested. Many of the impurities in detergent
preparations are not soluble in water; the cloudiness of a detergent solution at a concentration where it
is known to be soluble indicates the presence of an insoluble impurity.
Measurement of pH
The pH of the detergent solution is routinely measured. The pH should be neutral for detergents that are
either nonionic or zwitterionic.
Alcohol contamination
Glucoside and maltoside detergents are prepared from the corresponding hydrophobic alcohol. Trace
amounts of this alcohol in the detergent lot can cause cloudiness in a detergent solution. Therefore, we
measure the amount of alcohol in each lot of detergent by HPLC.
Alpha isomer
Glucoside and maltoside detergents have two isomeric forms - α and β. Each β detergent is analyzed for
the percent α isomer present by HPLC.
ANAPOE® detergents should be stored refrigerated in the dark. All other detergents should be stored frozen and
kept dry. Warm to room temperature before opening the container.

The lack of a complex glycerol ester chain and chiral centers allow these detergents to be prepared and
offered at moderate prices. FOS-CHOLINE®-12 (n-dodecylphosphocholine) micelles have proven to be
extremely useful as a medium in which to conduct NMR studies of membrane proteins1-4 and have also
been shown to play a crucial role in refolding a misfolded membrane protein5.
FOS-CHOLINE®-ISO-9, FOS-CHOLINE®-ISO-11
FOS-CHOLINE®-ISO-9 and FOS-CHOLINE®-ISO-11 possess split hydrophobic tails packing more hydrophobici-

NONIONIC DETERGENTS
Most of the detergents useful for the extraction and purification of membrane proteins are nonionic
detergents which possess either a sugar-based or a polyoxyethylene head group.
Sugar-based detergents
Sugar-based detergents are among the most popular detergents used for membrane protein applications.
The alkyl glucoside series is available from the six carbon to the twelve carbon tail. The somewhat less
polar thioglycoside analogs of some of these compounds are also available.

The high CMC value of hexyl−β−D-glucopyranoside (0.25M) indicates that a detergent with a shorter
chain would typically not be useful. Decyl−β−D-glucopyranoside has a reasonable CMC value, but is mar-
ginally soluble. Dodecyl−β−D-glucopyranoside is even less soluble.
For many membrane proteins, a member of the maltoside series is milder and more effective than the
comparable member of the glucoside series.

Another advantage of the maltoside head group is the increased solubility of the higher members of
this series compared to the glucoside series. Even hexadecyl−β−D-maltopyranoside is soluble although it
must be heated to 50oC for a few minutes to make a true solution. The entire series of maltoside deter-
gents is available from hexyl−β−D-maltopyranoside to hexadecyl−β−D-maltopyranoside, except for the
pentadecyl member.
Two sugar-based detergents that have been available for some time are sucrose monododecanoate and
sucrose monodecanoate. As with the maltose-based detergent series, these detergents are mild and use-
ful for the extraction of membrane proteins. However, these detergents are esters and thus are easily
hydrolyzed in acid, base or by proteases. The sucrose ester detergents are also mixtures of isomers in
which the aliphatic ester is linked to any one of the three primary alcohols in the sucrose structure.

The MEGA detergents are another series of glucose derivatives with an amide linkage between the alkyl
tail and the glycoside head group1-6. However, these detergents are poorly soluble in water.

CYMAL® Detergents
The CYMAL1,6 detergents are nonionic maltoside detergents that were designed specifically for the
extraction, purification and crystallization of membrane proteins2-4. These detergents have the formula:

The CMC values for CYMAL detergents are higher than for the corresponding linear chains containing
the same number of carbons. Viewed another way, the cyclohexyl tails pack more hydrophobicity into a
shorter effective chain length. It has been suggested that detergents that have a shorter chain length,
but higher hydrophobicity, are useful for stabilizing proteins during crystallization.
CYMAL-5® has also been shown to be effective for purifying G-protein coupled receptors5.

ANAPOE® Detergents
Most polyoxyethylene detergents used in the early studies of membrane proteins were developed for
large industrial applications. Today an increasing number of well defined, highly purified detergents,
designed specifically for the extraction of membrane proteins in their active state are available.
Detergents such as dodecyl−β−D-maltopyranoside and octyl−β−D-glucopyranoside have replaced many
polyoxyethylene detergents. Yet for some proteins, the polyoxyethylene detergents remain very useful—
particularly for routine extraction procedures.
Polyoxyethylene detergents are available in a variety of structures and under an often confusing list of
trade names, such as TRITON®, TWEEN®, GENAPOL®, BRIJ®, THESIT®, LUBROL®, etc. In addition to the confu-
sion caused by trade names, industrial detergents are often a mixture of closely related detergents that
may vary from lot to lot. They may also contain additives and contaminants which result in undesirable
effects during protein extraction. One such contaminant are peroxides, which can increase in concentra-
tion upon aging of the detergent solution.1
The “aging” of polyoxyethylene detergents results from the tendency of ethers to react with oxygen to
form peroxides; light accelerates this process. In samples of polyoxyethylene detergents, hydrogen perox-
ide and organic peroxides with a variety of structures may be found.2-4 In fact, the concentration of
hydrogen peroxide can be as high as 0.2%.5 Conversely, peroxides may react with detergent molecules in
solution, resulting in the presence of several undesirable derivatives.2
The presence of peroxides during extraction of membrane proteins can result in inactivation and/or
degradation of biological materials.4 Sulfhydryl groups are readily oxidized by peroxides and such oxida-
tion induces protein aggregation and inactivation6-8.

noted by Heath and Tappel when measuring lipid peroxides in detergent-solubilized membrane compo-

blue dye-binding assay, bicinchoninic acid method, and the Folin phenol method.10

MTS Reagents
The ability of MTS (methanethiosulfonate) reagents to react rapidly and specifically with sulfhydral
groups allows for the quantitative derivatization of transmembrane proteins at cysteine residues. A vari-
ety of sulfhydryl specific reagents can be generated by the modification of the functional group attached
to the methylthiosulfonate moiety. These various reagents can then be used to probe the structure or
topology of a protein. For example, charged MTS reagents are soluble in water and insoluble in a
hydrophobic environment. Therefore, reaction of charged MTS reagents with a cysteine residue indicates
that the specific cysteine group is probably on or near the membrane surface and not buried in the inte-
rior of the protein or membrane. Other MTS reagents contain a spin labeled or a fluorescent group to
probe the protein/membrane environment1-9.

NDSB Reagents
The series of NDSB reagents has been found to aid in the solubilization of some proteins. All members of
the series, which are zwitterionic over a pH range of 3-12 and are water soluble, have the unique ability
to reduce protein aggregation while stabilizing protein structure. Other applications include refolding
and renaturation of denatured proteins, isoelectrofocusing, and capillary electrophoresis. In addition,
NDSB compounds are of benefit in some protein crystallizations1-4.

To select the optimum detergent or combination of detergents and reagents for a particular membrane
protein application, oftentimes multiple detergents must be tested. Moreover, a detergent which is
suitable for extraction may not be useful for storage of the purified protein or for biochemical studies

DETERGENT SOLUBILITY
It is frustrating when one prepares a detergent solution that precipitates upon cooling to 4°C. There are
several possible reasons for such precipitation.
Combination of reagents causes precipitation
Precipitation can be caused by pH, ionic strength (salts), and the presence of organic solvent. Try making
a solution of each component to test its solubility in the relevant buffer.
Microbial growth causes precipitation
Alkyl glycoside detergents can be degraded by microorganisms, a process which can occur over a period
of days and which results in the appearance of a precipitate or suspension. Microbial growth can be sup-
pressed by including 1 mM EDTA in the buffer or by including a very low concentration of a bacteriocidal
agent such as sodium azide.
Presence of synthetic precursors
Alkyl glycosides and maltosides may contain small amounts of the parent alcohol, which may not be com-
pletely removed during purification. When a concentrated solution of detergent is prepared and cooled
to 4°C, a milkiness or cloudiness may become apparent due to precipitation by the alcohol. This can be
avoided by using highly purified detergents, such as ANAGRADE® detergents which contain less than
0.005% (w/w) of the parent alcohol, as compared with levels as high as 0.5% in some commercial prepa-
rations of the corresponding detergents.
Kinetic effect of detergent solubilization
Some detergents require heating for efficient solubilization. For example, when tetradecyl−β−D-maltoside
is dissolved at a concentration of 20% (w/v) at room temperature and cooled to 4°C, a precipitate appears.
However, if the detergent solution is heated to 50°C before cooling, the precipitate does not form.
Supersaturation
A concentration of detergent which is fully soluble at room temperature may be supersaturated at 4°C.
This is commonly observed for solutions containing sodium dodecyl sulfate. Supersaturated solutions can
exist for days, or even weeks, before precipitation occurs. The only recourse, when supersaturation is con-
firmed, is to reduce the detergent concentration or find an alternate detergent that is more soluble.

Detergent Grades
Generally, detergent purity is not as critical for extraction as for physiological studies and crystallization
processes. When selecting a grade of detergent for extraction, impurities that might degrade or modify
proteins should be avoided. The impurities of the maltosides and glucosides are generally the correspon-
ding alcohol of the tail portion of the detergent; these impurities are not reactive and therefore are
unlikely to harm the membrane protein being extracted. However, the presence of alcohol can lead to a
cloudy solution, particularly when the protein solution is cool.
For physiological studies and for crystallization of membrane proteins the purity of the detergents is
more important. Generally, the highest purity available should be utilized. In most instances, the deter-
gent of choice is a nonionic detergent. The purity should be listed as 99% or greater as measured by a
reliable test, such as HPLC. The conductance should be very low, indicating that there are few ionic impu-
rities. For those nonionic detergents without ultraviolet chromophores, both the ultraviolet absorbance
and the fluorescence background should be very low. Related detergents and aliphatic alcohols should be
low so as not to be detectable by HPLC or other reliable tests. It is also considered important by some
researchers that the detergent be isomerically homogenous. Thus, the aliphatic maltoside and glucoside
detergents should be 99% alpha or beta isomer.
For example, we offer three grades of Dodecyl maltoside, D310, D310A and D310S. D310S and D310A are
usually of sufficient purity for extraction and purification of membrane proteins. D310S is greater than
98% pure and often is more pure than the dodecyl maltoside available from other suppliers. D310A is
greater than 99% pure but contains up to 15% alpha isomer. D310 is 99% pure or greater and meets
many other purity requirements, such as low absorbance in the UV. This grade is generally used when
purification steps require high purity detergent or for studies using a purified membrane protein.
An additional grade (D310LA) contains less than 0.2% alpha isomer. Some researchers find this grade
preferable for crystallization studies. The alpha isomer itself is also available (D310HA).

Detergent Stability
The stability of a representative sample of detergents has been tested both in solution and in solid form.
The purpose of this study was to determine guidelines for shipping and storage of solid detergents and
to give some indication of their stability in solution.
To determine their stability during shipment, the following detergents were stored at 40°C for one week
and room temperature for two weeks: Dodecyl−β−D-maltoside, CYMAL®-3, HEGA®-10, MEGA-9,
Octyl−β−D-glucoside, CHAPS, CHAPSO, Dodecyl-N,N-dimethylglycine, Octyl−β−D-galactoside, Sodium
cholate, Dodecyl-N,N-dimethylamine oxide, and FOS-CHOLINE®-10. No deterioration was noted after this
time when the specifications for each sample were remeasured. That is, all detergent samples remained
within specifications.
To determine the stability of detergents in solution, a 10% solution of dodecyl maltoside was prepared and
incubated for 2 weeks at room temperature. The detergent specifications were re-measured including the
percent purity by HPLC, pH, conductance and absorbance. No deterioration was noted; the detergent
samples remained within specifications.
To determine the stability of detergents at elevated temperatures, 10% aqueous solutions of tetradecyl
maltoside, dodecyl maltoside, and HEGA®-10 were heated to 55°C for two hours. Re-measurement of the
specifications including the percent purity by HPLC, pH, conductance and absorbance indicated these
detergents had not deteriorated during this heat cycle.
