Why test for TOC ?

Quality & Reliability of scientific data depends on the

purity of the reagents used
Monitoring is required to warrant reagent’s purity
Water is the most commonly used reagent ; so far only the
ionic contamination has been monitored through
resistivity
Some laboratory applications are TOC sensitive (HPLC,
UPLC, ICP, LC, etc)
International norms and standards require low TOC values
for ultrapure water.

Standard Requirements
Standard specifications
 Explicit set of requirements for an item, material, component, system or service

Water standard requirements

 Several water standards are coexisting depending on the
– USP/EP: Purified Water, WFI (& Highly purified water)
– CLSI Reagent Water
– ASTM D1193 : Standard Specification for Reagent Water
– ISO 3696 : Water for analytical laboratory use

Guidelines
 Framework of common industry practices and recipes
– ISPE (International Society for Pharmaceutical Engineering)
– PIC/S pharmaceutical inspection convention/cooperation scheme
– CLSI - Clinical and laboratory Standard Institute
US and European Pharmacopoeia
CLSI* water quality specifications

Water qualities defined by the CLSI:

**Clinical laboratory Reagent Water (CLRW)

 CLRW Quantitative Specifications: Resistivity: ≥ 10 MΩ.cm referenced to 25° C; Total
Heterotrophic Plate Count: < 10 cfu/mL; TOC: < 500 ng/g (ppb); Particulates: puridication
stage that blocks the passage of particles ≥ 0.22 μm at or near the output stage.

Special Reagent Water (SRW)

Instrument Feed Water (IFW)
Water Supplied by a method manufacturer for use as a Diluent or Reagent
Commercial bottled, Purified Water
Autoclave and Wash Water Applications

*CLSI: Clinical and Laboratory Standards Institute (previously NCCLS)

** Concerning CLSI Water Qualities, complete CLSI corresponding sections should be read in order to understand the philosophy, method,
limitations, recommendations for each requirement.
