Chemistry
• Use a GNTT (green-top tube), an RTT (red-top tube) or a serum mini tube (gold-top tube).
Invert the GNTT 6–8 times immediately after adding blood and then spin down. Allow the
RTT or the gold-top serum mini tube to clot for 15–20 minutes before spinning.
• Use of a light GNTT (containing plasma-separator gel) is preferred for chemistries.
The light GNTT should be spun but does not require separation.
• When using a dark GNTT, an RTT or a gold-top serum mini tube, it is important
to separate the serum or plasma after spinning to avoid artifactual changes in
chemistry values.
Hematology
• For differentials, immediately make air-dried, unstained blood smears using glass slides.
Do not use coverslips. U se an individual slide holder for each patient.
• Submit blood in an EDTA lavender-top tube(LTT) or dark GNTT (without plasma-separator
gel). Invert tube 6–8 times immediately after adding blood. Minimum blood volume in
GNTT/LTT should be ≥0.3 mL.
• DO NOT use an LTT with reptile, kookaburra, crow, ostrich or emu specimens.
Microbiology
In psittacine or passerine avian microbiology, organisms are categorized as nonpathogenic
(reported as normal Gram-positive flora), including a-hemolytic streptococci (alpha-strep)
and g-hemolytic streptococci (gamma-strep), Enterococcus, Corynebacterium, Lactobacillus,
nonhemolytic staphylococci; or pathogenic, including Enterobacteriaceae and other
Gram-negative rods, fungi, Mycoplasma, hemolytic staphylococci, Pasteurella and yeast on
respiratory and fecal samples. Yeast are noted as present but are not identified.
Susceptibility testing is routinely performed on all Enterobacteriaceae, Gram-negative rods
and hemolytic staphylococci. Pasteurella is NOT tested because it is routinely susceptible to
all avian antibiotics.
Note: Antibiotics tested for avians are dependent on the organism and may include amikacin,
Specimen Collection
Pathology
IDEXX Reference Laboratories has a team of clinical and anatomic pathologists with special
expertise in avian and exotic species. If you would prefer your biopsy, cytology or other
relevant testing to be read by a member of this team, please specify “Direct case to exotics
pathologist” on your requisition form. Note that this may result in a slightly longer turnaround
time to allow for routing of your case to a particular pathologist.
IMPORTANT: Always prioritize tests on your requisition form in case the specimen
size is inadequate.

animals). Avoid contamination with feces.
Swabs are not acceptable specimens for
semiquantitative urine culture.
*An LTT is not recommended for cultures as it may inhibit bacterial growth. An SST (serum separator tube) is not appropriate
for cultures. A urine culture tube with boric acid is only recommended for use when submission of urine may be significantly
delayed. Do not underfill urine culture tubes.
Special instructions
• Inform the laboratory if the animal is being treated with antimicrobial drugs.
• Indicate if the patient is immunocompromised.
• DO NOT submit syringes with needles.
Specimen Collection
• Clearly specify sites of all cultures, including method of collection for urine specimens.
• Urine, fluids, aspirates and tissues may be submitted in a WTT (plain plastic tube) or an
RTT. An LTT is not recommended for cultures as it may inhibit bacterial growth. An SST
(serum separator tube) is not appropriate for cultures.
• Culture specimens should be refrigerated promptly to prevent overgrowth, with the
exception of blood cultures, CSF and joint fluid, which should be kept at room temperature.
• Recent antibiotic therapy (within the previous 2 weeks) may result in lower yield or
negative cultures. Withdrawal from antibiotics for a minimum of 72 hours, ideally
7–10 days, is recommended when culturing a patient following antibiotic administration.
• When submitting two swabs from different sites, submit each swab with its own
requisition form to be plated separately. If you would prefer these swabs to be cultured
together on the same plate under a single test code, submit together and indicate this on
the requisition form.

specimens (formalin-fixed, paraffin-embedded tissue). Fresh, refrigerated tissue specimens
submitted within 24 hours of collection are also acceptable. Freezing is not recommended
unless the specimen can be maintained frozen until arrival at the PCR laboratory.

What Is an MIC?
The MIC, or minimum inhibitory concentration, is the lowest concentration (in μg/mL) of
an antibiotic that inhibits the growth of a given strain of bacteria. At IDEXX, a commercial
automated system is used to determine MICs. A quantitative method of susceptibility testing,
an MIC helps determine which class
of antibiotic is most effective. This Culture result, susceptibility

Antibiotics
When selecting an antibiotic, keep in mind that other factors in addition to the MIC are
important. The location of the infection is important because lipid-soluble drugs reach
higher levels in the tissue than they do in serum. Drugs excreted by the kidney reach much
higher bladder levels than serum levels. Also, some drugs are more effective against
Gram-negative bacteria than Gram-positive bacteria and vice versa. Species considerations
are also important because certain antibiotics are toxic in some species.

A small study suggests that a short course (2 weeks) of Apoquel® likely does not interfere
with serum allergy testing. Studies looking at the effects of longer term use of Apoquel are
not yet available. It may be ideal to consider discontinuation of Apoquel for at least 4–12
weeks prior to testing if an animal has been on this medication long-term.
Immunotherapy Options
Along with your patient’s serum allergen-specific IgE panel results, you will be provided a
letter with guidance for ordering the appropriate GREER® Aller-g-complete® immunotherapy
based on the results and owner preference for delivery.
GREER® Aller-g-complete® now has two immunotherapy options for your atopic patients,
either subcutaneous or sublingual. The new sublingual immunotherapy, GREER® Aller-g-
complete® Drops, may help increase compliance for some pet owners because of the ease
of oral administration. This therapy option is new to veterinary medicine, and studies have
shown that it is a safe alternative to subcutaneous immunotherapy.

For most atopic patients, ordering the Regional Allergen Panel is indicated. This panel
tests for IgE against 36 core environmental allergens common to all regions and
12 additional allergens specific to your region. These 48 allergens include pollens (from
weeds, trees and grasses), molds and mites (including storage mites often found in pet
food). If the patient lives near the border of a region, an Add-on Secondary Regional
Allergen Panel from the neighboring region may also be indicated.
Additional panels that are less commonly indicated, but can be ordered in combination
with the Regional Allergen Panel, include the Farm & Stable Panel (for common
farm allergens, such as alfalfa, oats etc.), Food Panel and Insect Panel (for common
household and biting insects).

Send-out Tests
There are some esoteric tests that are not performed by an IDEXX reference laboratory.
As an added service, IDEXX will send many of these tests to specialized or university
laboratories. Turnaround times listed for these tests are subject to change according to the
individual laboratory that runs the test.
ASAP Turnaround
Check the ASAP box on your test requisition form, and if feasible, we’ll give your specimens
priority during the regular run. Please reserve this option for critical patients as it may cause
delays in processing of specimens from other patients.
