“ESTIMATION OF TANNIN METABOLITES IN

BLOOD SERUM AND COW MILK USING HPLC.”

This method described the estimation of different monomers of tannin in serum and cow
milk. Animals were supplemented with two types of diets containing 0 and 4 per cent
tannin (DM basis) from Acacia nilotica pods. Blood and Milk samples were collected
after a preliminary feeding of two and half months and were processed for the estimation
of degraded products of babul pods tannin. Phloroglucinol, resorcinol, catechin,
epicatechin and catechin gallate were identified and quantified in blood serum sample
and the values of various tannin monomers appeared were 128.07.0.17, 2.29, 63.65, and
51.17 ug/dl respectively, while in milk the values were 268.26,2.89, 298.19 and 2.14
ug/dl for phloroglucinol, resorcinol, gallic acid and catechin respectively. Thus, it is
indicative of the fact that catechin gallate available from the serum is further metabolized
into catechin and gallic acid as end products in the mammary gland.

This method described the estimation of different monomers of tannin in faeces and urine
or cows. Animals were supplemented with two types of concentrate containing 0 and 4
per cent tannin (DM basis) from Acacia nilotica pods. Faeces and urine samples were
collected after a preliminary feeding of two and half months period and were processed
for estimation of degraded products of tannin from Acacia nilotica pods. Phloroglucinol,
gallic acid and catechine were identified and quantified in the faeces and the values were
4.74, 0.45 and 0.71ug/g tannin respectively, where as in the urine of cows phloroglucinol,
gallic acid, resorcinol, catechin, epicatechin and catechin gallate were identified and
quantified and the values were 268.0, 100, 610, 60.0, 80 and 206.0 ug/g tannin
respectively.

This method described the estimation of different tannin monomers present in feed
samples. Feed were hydrolysed with 2M sulphuric acid at 100 degree C. for 24h to get
the tannin monomers. Tannin monomers were identified and quantified in filtrate after
filtration of hydrolysed sample through grade 1 filter paper followed by 0.45m Millipore
filter paper. The presence of tannin monomers in acacia nilotica (pods). Mangifera indica
(kernel), shorea robusta (cake) and Madhuca indica (cake) were invented using the above
method. All the samples produced gallic acid upon acid hydrolysis. Phloroglucinol was
produced only in the case of shorea robusta. Catechin was produced by both shorea
robusta and Mangifera indica. Epicatechin was produced in Madhuca indica. None of the
condensed tannin hydrolyzed products e.g. catechin and epicatechin were detected in
acacia nilotica pods. Presence of tannin monomers in above feeds were identified and
estimated.
