Using Lactose Monensin Glucuronate (LMG) Agar, the ISO-GRID and
NEO-GRID Membrane Filter Systems are used to provide a method Monensin 0.038 g/L
for detecting and confirming a sample’s coliform and E. coli count. T7 0.15 g/L
Buffered MUG Agar (BMA) is used for E. coli confirmation. Aniline Blue (CI #42755) 0.1 g/L

The ISO-GRID/NEO-GRID systems are based on the principle of Final pH: 7.2 ± 0.2
hydrophobic grid membrane filtration. Coliforms and E. coli are de-
tected through the use of a unique membrane filter containing a grid Buffered MUG Agar (BMA)
of 1,600 squares. First, a diluted sample is prefiltered to eliminate any Dibasic Sodium Phosphate 8.23 g/L
food particles present. The sample is then filtered through a hydrophobic Monobasic Sodium Phosphate 1.20 g/L
membrane, and the membrane is placed on LMG Agar. LMG Agar is Sodium Chloride 5 g/L
specifically formulated to detect coliforms. 4-Methylumbelliferyl-B-D-glucuronide 0.1 g/L (MUG)
After incubation, the membrane filter is examined for the presence Agar 15 g/L
of blue colonies. Any blue (presumptive coliform) colonies present Final pH: 7.2 ± 0.2
are confirmed using procedures detailed in the Methods Manual. To

Test Procedure For Use With Either the NEO-GRID or ISO-GRID Test System
6902A LMG Agar
1. Prepare a sample homogenate. 6903A BMA Agar
2. Filter 1.0 mL of homogenate through an ISO-GRID or 6810 1-Place Filtration Manifold
NEO-GRID hydrophobic grid membrane filter. 6800 3-Place Filtration Manifold
3. Place the membrane filter on Lactose Monensin Glucuronate 6801 6-Place Filtration Manifold
(LMG) Agar. 6821 Rubber Tubing (10 feet)
4. Incubate for 24 ± 2 hours at 35–37°C. Obtain coliform count. 6822 Side-Arm Flask Assembly (1 L)
5. Transfer membrane filter to BMA. 6824 Vacuum Pump - ¼ hp (Gast)
6. Incubate BMA plates for 2 hours at 35–37°C. 6828 Forceps
