ULTRAVOILET SPECTROPHOTOMETER
• When a beam of radiant energy strikes the surface of a
substance, the radiation interacts with the atoms and
molecules of the substance.

The radiation may be


• Transmitted
• Absorbed
• Scattered
• Reflected depending upon the properties of the
substance.

PRINCIPLE

• It is based upon the principle that the amount of


absorption that occurs is dependent on the number of
molecules present in the material.

CONSTRUCTION & WORKING
• The ultraviolet spectrometer consists of a radiant source
of hydrogen or deuterium source.
• The radiation from the source through the condensating
lens is made to fall on the focusing lens through the
entrance slit.
• The radiation from the light source is passed either
directly by means of a lens or mirror into the narrow slit
of the monochromator and allowed to fall on the
dispersing medium, where it gets isolated.

• Here the dispersing medium is the diffraction grating.
• The isolated radiation is passed through the cuvette
where the sample is held. The radiation may be
absorbed, scattered or reflected by the sample.
• This isolated radiation of a particular wavelength is
selected by a proper filter.

• This radiation is made to fall upon a photo sensitive
element in which the light energy is converted into
electrical energy.

• The electric current produced can be measured with a


sensitive galvanometer directly or after suitable
amplification.

Radiation source

• The ultraviolet radiation ranges from 200nm to 400nm.


The radiation sources used for ultraviolet region are
hydrogen lamp and deuterium lamp

Monochromator

• For better isolation of spectral energy diffraction grating


is used for UV spectrometer.

Optical components

• The optical components in the UV spectrophotometer


are entrance slit, exit slit and condensating lens.

Detector

• The detectors used for a UV spectrometer are photocells


and photo multiplier tubes.

determination of solutions of transition metal ions and
