Psychiatric diseases are the main cause of disability around the world. In
spite of the high economic and social burden, the development of new
treatments has been slow. The deep brain stimulation (DBS) is a
surgically alternative for the modulation of deep central nervous
structures, which has been widely used in the treatment of Parkinson’s
disease symptoms through the stimulation of the subthalamic nucleus and
internal globus pallidus internus. The DBS of the amygdala, a key
structure associated with the defensive system, has recently been
proposed in the treatment of stress and anxiety disorders. The
development of more specific and efficient stimulation protocols is one
of the main necessities regarding the use of DBS in neuropsychiatry.
Given so, the present study investigated the acute unilateral amygdala
DBS effects using two frequency different bands (130 and 20 Hz) and
three diverse current amplitudes (10, 50 e 100 µA) over rats’ behavior.
Under the experimental conditions used, neither defensive behavior nor
fear memory consolidation were affected by acute stimulation. As
amygdala DBS is a potential therapeutic approach for the treatment of
stress and anxiety disorders, these data contributes in a translational
fashion for the improvement of the technique.

1 INTRODUÇÃO

Compreende-se que a tradução literal do termo seria “Estimulação Encefálica
Profunda”. No entanto, faremos uso do termo na forma como está traduzido em
publicações de referência nacionais (DIAZ et al., 2015)

1.1 JUSTIFICATIVA
1.2 HIPÓTESE
Estudos prévios demonstraram que a expressão de comportamento
tipo-hipervigilante (LANGEVIN et al., 2010; STIDD et al., 2013) e a
evocação da memória aversiva (SUI et al., 2014) foi atenuada em ratos
tratados cronicamente com ECP da amígdala. Em contrapartida, a
expressão de comportamentos defensivos não seria afetada por meio da
estimulação crônica (STIDD et al., 2013), ainda que tenha sido modulada
por meio da estimulação super-aguda (1 s) da mesma estrutura em estudo
anterior (SALDÍVAR-GONZÁLEZ et al., 2003). Com exceção do último
estudo (SALDÍVAR-GONZÁLEZ et al., 2003), os demais utilizaram
estimulação monopolar e unilateral direita de alta frequência (acima de
100 Hz) (LANGEVIN et al., 2010; STIDD et al., 2013; SUI et al., 2014)
A ECP da amígdala pode ter aplicabilidade terapêutica no caso de
comportamentos sabidamente patológicos e que afetam a qualidade de
vida de pacientes. Por outro lado, a ECP desta estrutura límbica poderia
ter como efeito colateral a interferência sobre respostas fisiológicas e
adaptativas com componente emocional relevantes para o desempenho
cognitivo e, até mesmo do ponto de vista evolutivo, importantes para a
preservação da espécie. Desta forma, nossa hipótese é que a estimulação
unilateral de alta frequência da amígdala de ratos poderia reduzir a
expressão de comportamentos defensivos bem como prejudicar a
consolidação da memória aversiva nestes animais.
