Contudo, desenvolvem padro axial com os rgos mais ou menos na posio correcta e diferenciam todos os tipos de tecidos em disposio radial. Sabe-se que os mutantes tm nveis elevados de IAA que pode originar o crescimento anormal. (O aumento de IAA pode dever-se a uma deficincia na enzima que normalmente inactiva a fito-hormona, conjugando o IAA livre com amino cidos). 2 Estas mutaes, s/ banda pr- profsica e com divises celulares aberrantes, no impedem completamente o estabelecimento do padro radial. Poderia inferir-se que o controlo preciso e ordenado do plano de diviso celular no essencial para o estabelecimento do padro radial de diferenciao histolgica. 2 Mutaes knolle em Arabidopsis Mutantes com deficiente citocinese por inactivao do gene que codifica uma protena semelhantea uma sintaxina importantena Mutantes com citocinese irregular no estabelecem o padro radial semelhante a uma sintaxina importante na fuso de vesculas. (Sintaxinas - protenas integradas nas membranas que permitem a fuso membranar). A diviso nuclear no bloqueada, contudo, a placa celular irregular e incompletamuitas clulas binucleadas, outras parcialmente separadas. Os planos de diviso, tambm, so irregulares acarretando efeitos severos no desenvolvimento. Mutao knolle no impede a formao do eixo d l id d ( i l b l) b i 3 de polaridade (apical-basal) e a embriognese completa-se; os mutantes homozigticos completam o desenvolvimento embrionrio, mas a disposio radial dos tecidos severamente alterada, no se forma a camada protodrmicanas fases iniciais da embriognese. Contudo, ton diferente de knolle os mutantes ton estabelecem o padro histolgico radial a mutao knolle impede o estabelecimento do padro histolgico radial porque no permite a separao efectiva das clulas filhas durante a citocinese uma vez que a placa celular fica incompleta. Ou seja, nos mutantes ton as clulas so capazes de detectar a informao posicional enquanto que nos mutantes knolle as clulas no detectaminformao posicional. 4 A formao de um padro est muito dependente de informao posicional pelo que necessrio que as clulas estejam efectivamente separadas para que a informao posicional possa ser regulada. Em todos os organismos pluricelulares a especificao do destino celular e a coordenao do desenvolvimento dependem da comunicao entre clulas. Uma das formas de comunicao das clulas vegetais consiste no trnsito selectivo de factores de transcrio e de outros sinais atravs dos plasmodsmios. 3 Sinais intercelulares no desenvolvimento da raz (cont.) Estudos de ablao por laser (Van den Berg et al, 1995) Aps ablao, o curso de desenvolvimento das clulas foi analisado atravs da expresso de genes marcadores especficos de determinados tecidos. Alguma informao determinante do padro radial parece originar-se em segmentos mais velhos da raiz. 7 Se se destrurem 3 clulas derivadas no sentido radial o efeito diferente. Destruindo uma clula derivada da cortical inicial juntamente com a clula de pericclo adjacente mais a clula epidrmica no exterior, o desenvolvimento da raiz no afectado. A inicial cortical por baixo da clula destruda forma uma nova derivada que se divide assimetricamente. Estes trabalhos sugerem que a informao do posicionamento transmitida dos tecidos mais velhos para o pice da raiz. Informao posicional determina o destino A alterao do destino de clulas que mudam de camadas implica que a informao posicional (e no determinantes baseados na linhagem) que controla a aquisio, pelas iniciais meristemticas e suas derivadas, das caractersticas tpicas dos tecidos. O padro histolgico da raiz - estando a arquitectura inicial estabelecida pelos genes do padro embrionrio resulta em parte, da especificao histolgica de clulas em desenvolvimento, atravs de sinais provenientes de clulas mais desenvolvidas i e 8 atravs de sinais provenientes de clulas mais desenvolvidas i.e., dos tecidos adultos. 5 Concluses A informao posicional desempenha umpapel A informao posicional desempenha um papel fundamental no controlo da identidade das clulas e tecidos. Posicionamento do meristema apical caulinar (SAM) (adiante) Actividade ontognica dos Meristemas Apicais O desenvolvimento das plantas ocorre na sua maioria aps embriognese (durante a embriognese estabelece-se somente uma estrutura axial rudimentar, com os pices caulinar e radicular nas extremidades). Os meristemas dos pices caulinar e radicular (estabelecidos durante a embriognese) s entram em actividade aps a embriognese estar completa e a germinao se iniciar. Os meristemas apicais so constituidos por clulas com capacidade de se dividir tanto quanto durar o crescimento, i.e., teoricamente podem dividir-se durante toda a vida da planta. Deste modo, nos meristemas apicais ocorre embriognese contnua e permanente na sequncia da segmentao do zigoto e da edificao intraseminal dos primeiros rgos do embrio.
