Influence of maternal thyroid hormones during gestation on fetal brain
development.

Abstract

Thyroid hormones (THs) play an obligatory role in many fundamental processes underlying brain development and maturation. The
developing embryo/fetus is dependent on maternal supply of TH. The fetal thyroid gland does not commence TH synthesis until mid gestation,
and the adverse consequences of severe maternal TH deficiency on offspring neurodevelopment are well established. Recent evidence
suggests that even more moderate forms of maternal thyroid dysfunction, particularly during early gestation, may have a long-lasting influence
on child cognitive development and risk of neurodevelopmental disorders. Moreover, these observed alterations appear to be largely
irreversible after birth. It is, therefore, important to gain a better understanding of the role of maternal thyroid dysfunction on offspring
neurodevelopment in terms of the nature, magnitude, time-specificity, and context-specificity of its effects. With respect to the issue of context
specificity, it is possible that maternal stress and stress-related biological processes during pregnancy may modulate maternal thyroid
function. The possibility of an interaction between the thyroid and stress systems in the context of fetal brain development has, however, not
been addressed to date. We begin this review with a brief overview of TH biology during pregnancy and a summary of the literature on its
effect on the developing brain. Next, we consider and discuss whether and how processes related to maternal stress and stress biology may
interact with and modify the effects of maternal thyroid function on offspring brain development. We synthesize several research areas and
identify important knowledge gaps that may warrant further study. The scientific and public health relevance of this review relates to achieving
a better understanding of the timing, mechanisms and contexts of thyroid programing of brain development, with implications for early
identification of risk, primary prevention and intervention.

GABA

• GABA is an inhibitory neurotransmitter that is often referred to as “nature’s


VALIUM-like substance”. When GABA is out of range (high or low excretion
values), it is likely that an excitatory neurotransmitter is firing too often in the
brain. GABA will be secreted to attempt to balance this stimulating, excessive,
over-firing

• Glutamine is transformed into GABA (highlighted in blood group O)

• DOPAMINE is a special neurotransmitter because it is considered to be both


excitatory and inhibitory

• Dopamine helps with depression as well as focus

• Excitatory Neurotransmitters - DOPAMINE is our main focus neurotransmitter. When
dopamine is either elevated or low – we can have focus issues such as not remembering
where we put our keys, forgetting what a paragraph said when we just finished reading it or
simply daydreaming and not being able to stay on task. Dopamine is also responsible for our
drive or desire to get things done – or motivation. Stimulants such as medications for ADD/
ADHD and caffeine cause dopamine to be pushed into the synapse so that focus is
improved. Unfortunately, stimulating dopamine consistently can cause a depletion of
dopamine over time. Noradrenaline is an excitatory neurotransmitter that is responsible for
stimulatory processes in the body. Noradrenaline helps to make adrenaline as well. This
neurotransmitter can cause ANXIETY at elevated excretion levels as well as some “MOOD
DAMPENING” effects. Low levels of noradrenaline are associated with LOW ENERGY,
DECREASED FOCUS ability and sleep cycle problems. Adrenaline is an excitatory
neurotransmitter that is reflective of stress. This neurotransmitter will often be elevated
when ADHD like symptoms are present. Long term STRESS or INSOMNIA can cause
adrenaline levels to be depleted (low). Adrenaline also regulates HEART RATE and BLOOD
PRESSURE

In Vitro TNF-α Inhibitory Activity of Brazilian Plants and Anti-Inflammatory Effect


of Stryphnodendron adstringens in an Acute Arthritis Model.

Abstract

Stryphnodendron species, popularly named "barbatimão," are traditionally used in Brazil as anti-inflammatory agents. This study aimed to investigate the effect of barbatimão and 11 other species on
the production of tumor necrosis factor-alpha (TNF-α) in lipopolysaccharide- (LPS-) stimulated THP-1 cells, as well as their anti-arthritis activity. The extracts of Stryphnodendron
adstringens, Stryphnodendron obovatum, Campomanesia lineatifolia, and Terminalia glabrescens promoted a concentration-dependent inhibition of TNF-α. Mice injected with LPS in the knee joint were
treated per os with fractions from the selected extracts. Both the organic (SAO) and the aqueous (SAA) fractions of S. adstringens promoted a dose-dependent reduction of leukocyte migration and
neutrophil accumulation into the joint, but none of them reduced CXCL1 concentration in the periarticular tissue. In contrast, treatment with C. lineatifolia and T. glabrescens fractions did not ameliorate
the inflammatory parameters. Analyses of SAO by Ultra Performance Liquid Chromatography (UPLC) coupled to electrospray ionization mass spectrometry (ESI-MS) led to the identification of gallic
acid along with 11 prodelphinidins, characterized as monomers and dimers of the B-type. Our findings contribute to some extent to corroborating the traditional use of S. adstringens as an anti-
inflammatory agent. This activity is probably related to a decrease of leukocyte migration into the inflammatory site. Polyphenols like gallic acid and prodelphinidins, identified in the active fraction, may
contribute to the observed activity.

Evaluation of the Effects of Some Brazilian Medicinal Plants on the Production of TNF- α and CCL2 by THP-1 Cells.

Abstract

Several plant species are traditionally used in Brazil to treat various inflammatory diseases. Tumor necrosis factor- (TNF-) α and chemokine (C-C motif) ligand 2 (CCL2) are key inflammatory mediators
in diseases like rheumatoid arthritis and atherosclerosis, respectively; nevertheless, only a few extracts have been assayed against these targets. We herein report the effect of 19 plant extracts on TNF-
α and CCL2 release by lipopolysaccharide- (LPS-) stimulated THP-1 cells, a human monocytic leukemia cell line, along with their radical scavenging activity on DPPH. The extracts of Caryocar
brasiliense, Casearia sylvestris, Coccoloba cereifera, and Terminalia glabrescens inhibited TNF-α production in a concentration-dependent manner. Fractionation of these extracts potentiated the anti-
TNF-α effect, which was shown to concentrate in polar fractions, mainly composed by polyphenols. Significant CCL2 inhibition was elicited by Lippia sidoides and Terminalia glabrescens extracts,
whose fractionation resulted in highly active low polar fractions. All assayed extracts showed strong radical scavenging activity, but antioxidant activity did not correlate with inhibition of TNF-α or CCL2
production. Our results allowed identifying extracts with selective capacity to block cytokine production; therefore, further purification of these extracts may yield molecules that could be useful in the
treatment of chronic inflammatory diseases.

Abstract

For several years, reproductive immunology has been dominated by the 'Th1/Th2' hypothesis, in which the fetus avoids
maternal T-cell rejection through a bias towards T-helper (Th)2 cytokine production. The discovery that normal pregnancy
is a controlled state of inflammation, at an early stage at the implantation site and also later systemically, has challenged
this concept, as has the finding that the predominant immune interactions in the decidua are between the placental
trophoblast and maternal natural killer (NK) cells instead of T cells. Here, we extend this concept to the interaction
between the trophoblast and NK cells in the maternal circulation. We suggest novel ways in which the trophoblast might
stimulate the maternal systemic inflammatory response, and how dysfunctional NK-cell activation could result in the
maternal syndrome of pre-eclampsia.
