Os efeitos do processamento sobre a composição metabólica química, físico-química,
enzimática e volátil da polpa de pitaya foram avaliados pela primeira vez. Para tanto, foram
avaliados os seguintes tratamentos para obtenção da polpa de pitaya: Tratamento A (AT,
processamento de polpa sem ácido ascórbico), Tratamento B (TB, processamento de frutas

O tratamento empregado naExportação
TB resultou em baixa atividade de polifenol oxidase e
peroxidase e nenhuma alteração química ou físico-química significativa na maioria dos
parâmetros avaliados. Além disso, a TB apresenta altos rendimentos e teor de fibras em
comparação com o AT ou Controle. Para análise metabólica, a Cromatografia Gasosa-
Espectrometria de Massa (GC-MS) foi eficaz para a determinação simultânea de 80
metabólitos voláteis em pitaya.

1 . Introdução
Pitahaya, pitaya e fruta do dragão ( Hylocereus spp.) São nomes comuns para um grupo
diversificado de gêneros da família Cactaceae. As três espécies mais comuns de pitaya
comercializadas são as pitaya com pele vermelha e carne branca ( Hylocereus undatus Haw.
Britton & Rose), pele vermelha e carne vermelha ( Hylocereus sp.) E pele vermelha e carne
vermelha forte ( Hylocereus polyrhizus Webb. Britton Rose). Devido à sua cor e sabor atraentes, a
produção mundial está aumentando rapidamente e novas culturas de frutas para áreas secas
estão em desenvolvimento ( Mercado-Silva, 2018 ).
Pitaya é nativa da América Central e do Sul ( Mercado-Silva, 2018 , Ortiz Hernandez e Carrillo-
Salazar, 2012 ) e é cultivada comercialmente em uma ampla variedade de países, incluindo

semiárida do Brasil, como no Ceará ( Nunes et al., 2014 ). Uma
vantagem particular do cultivo de pitayas como uma colheita em regiões secas é o seu
potencial de contribuir para o desenvolvimento agrícola nessas regiões em virtude de sua alta
tolerância ao estresse de seca a longo prazo e solos pobres ( Hua et al., 2018 , Wu et al. , 2019 ).
Nos últimos anos, a pitaya ganhou popularidade, pelo menos em parte, como resultado da
aparência atraente da carne e devido aos potenciais benefícios à saúde dos altos níveis de
betalaína, característicos das variedades de pitaya vermelhas ou roxas ( Rodriguez- Amaya,
2019 , Stintzing e Carle, 2007 , Wybraniec e Mizrahi, 2002 ). A fonte mais comum de betalaína é
a beterraba vermelha ( Beta vulgaris L. ssp. Vulgaris), porém apresenta altos níveis de nitrato (
Belhadj Slimen et al., 2017 , Santamaria, 2006 ) e um cheiro de terra causado por derivados de
geosmina e pirazina ( Lu Edwards, Fellmann, Mattinson e Navazio, 2003) Na busca de novas
plantas contendo betalaína sem essas desvantagens, o pitaya surge como uma alternativa
interessante. Nesse sentido, estudos que descrevem o processamento de pitaya em sucos de
frutas e produtos relacionados foram intensificados nos últimos anos.

2.1 . Matéria-prima

Pitaya ( Hylocereus polyrhizus (FAC Weber) Britton & Rose) foi fornecida pela Frutacor, localizada
em Vale do Jaguaribe - CE (05 ° 53′26 ″ S; 38 ° 37′19 ″ W). A fruta foi lavada, higienizada e
processada no Laboratório de Processos Agroindustriais da Embrapa Agroindústria Tropical
(Fortaleza-CE).
2.2 . Reagentes
Catecol (1,2-di-hidroxibenzeno), polivinilpirrolidona (PVP) e polivinilpolipirrolidona (PVPP)
foram adquiridos na Sigma-Aldrich (St. Louis, MO, EUA). Os reagentes fosfato de potássio,
peróxido de hidrogênio e bissulfito de sódio foram obtidos da Vetec Quimica Fina Ltd.
(Duque de Caxias, Brasil). Todos os produtos químicos eram de grau analítico ou superior
(Sigma-Aldrich Canada Ltd., Oakville, Canadá).

As frações insolúveis (IDE) e solúveis (SDF) da fibra alimentar foram determinadas de acordo

com o método AOAC Compartilhar Exportação
991.43 ( AOAC, 2005 ) utilizando um analisador ANKOM Automatic
Nutrient TDF (Ankon Technology Corporation). A fibra alimentar total (TDF) foi calculada
como a soma das frações insolúveis e solúveis. Além disso, os sólidos solúveis (° Brix) foram
avaliados usando um modelo de refratômetro digital (ATAGO Pocket Palm-3). O pH e a acidez
titulável foram avaliados utilizando um potenciômetro de titulação (HI 901 / HI 902, Hanna
Instruments) de acordo com a Association of Official Analytical Chemistry, 2005 , Instituto
Adolfo Lutz, 2008 , respectivamente.
2.6 . Atividade da cor, polifenol oxidase (PPO) e peroxidase (POD) da polpa de pitaya
Para avaliar a atividade enzimática da polifenol oxidase (PPO) e peroxidase (POD) da polpa de
pitaya (TA, TB e Controle), foi preparado um extrato enzimático de cada material. Para a PPO,
foi utilizado o método descrito por Sojo, Nunez-Delicado, Garcia-Carmona e Sanchez-Ferrer
(1998) , com adaptações. Resumidamente, em um banho de água gelada, 0,1 de polpa foi
homogeneizada por 5 min com 10 mL de tampão KH 2 PO 4 (0,01 mol L -1 , pH 6,0) contendo
4% 0,04 mg g -1 Triton X-100 e 0,02 g de polivinilpolipirrolidona (PVPP). Após centrifugação
(5000 gpor 40 min a 4 ° C), o precipitado foi descartado e o sobrenadante foi incubado em
banho-maria por 10 min (37 ° C). O sobrenadante foi então centrifugado por 20 min (2000 g , 4
° C) e usado como um extrato para a análise da atividade da PPO, usando o método
estabelecido por Robinson (1987) . Neste método, a mistura de reação consistiu em 80 μL de
tampão fosfato de potássio (0,1 mol L -1 , pH 6,0), 100 μL de extrato e 5 μL de substrato de
pirocatecol (0,1 mol L −1 ). Após incubação a 30 ° C por 30 min, a absorbância foi medida a 395
nm. Uma unidade de atividade enzimática (em unidades de atividade, UA) foi definida como
uma variação de 0,001 na absorvância. Os resultados foram expressos em UA / min.mg.P e
calculados considerando o controle como 100%.

O extrato enzimático para a determinação do POD foi preparado de acordo com o método

e Lee (1980) , com algumas modificações. Em um banho de água
gelada, 0,5 g de polpa foi homogeneizado com 10 mL de KH 2 PO 4 tamp (0,05 mol L -1 , pH
7,0) contendo 1% de polivinilpirrolidona (PVP) durante 3 min. Após centrifugação (10.000 g
por 10 min a 4 ° C), o precipitado foi descartado e o sobrenadante foi considerado a fonte
enzimática. O sobrenadante foi usado como um extrato para a análise da atividade do POD. A
atividade foi determinada de acordo com o método descrito por Matsuno e Uritani (1972) . A
mistura de reação consistiu em 55 μL de KH 2 PO4 tampões (0,05 mol L -1 , pH 7,0), 300 μL do
extrato e 55 μL de 3% de peróxido de hidrogênio. Após incubação a 30 ° C por 5 min, a reação
foi extinta com a adição de bissulfito de sódio a 30%. A absorvância foi então medida a 470 nm
e os resultados foram expressos em UA / min.mg.P. Os resultados foram calculados
considerando o controle como 100%.

Os dados das áreas de pico dos compostos voláteis, obtidos por cromatogramas, foram
normalizados pela mediana tratada na escala logarítmica e escalonados de acordo com a escala
de Pareto. A análise quimiométrica usando o Metaboanalyst 3.0 ( www.metaboanalyst.ca ) foi
realizada com base nas matrizes de correlação anormalizadas dos dados normalizados. Para
análise de variância (ANOVA), LSD (teste de menor diferença significativa) de Fisher, análise de
FDR, agrupamento hierárquico e análise de mapa de calor, foi utilizado o programa Minitab
17 ( Xia, Sinelnikov, Han & Wishart, 2015 ).

Para proteínas e lipídios, os valores foram inferiores a 0,5%. De acordo com nossos dados,
Abreu et al., 2012 , Sato et al., 2014 também relataram baixos níveis de proteínas e lipídios na
polpa de pitaya. Os resultados obtidos com a casca de pitaya não apresentaram alterações em
nenhum dos parâmetros avaliados, exceto no que se refere à umidade e ao teor de
carboidratos ( Tabela 1 ). A casca obtida pela TB apresentou baixos valores de umidade, o que
explica o uso de toda a pitaya no processo de polpação. O finalizador foi utilizado para
pressionar o material e, consequentemente, remover a polpa aderida à casca. Como resultado,
foi observada uma redução na umidade. Da mesma forma, o teor de fibras na TB foi maior
que no AT, devido ao tipo de processamento empregado (processamento de frutas inteiras).

Treatment A (TA): pulp processing without ascorbic acid, and Treatment B (TB): whole fruit processing with
ascorbic acid, and Control: whole fruit processing without ascorbic acid. Results expressed in mean and
standard deviation; Averages with equal letters, in the same line, do not differ at the level of 5% of
significance for the Tukey test.
The PPO and POD activity was significantly higher in the Control, confirming that in whole
fruit processing, the compounds present in the peel were carried out to the pulp, improving
its enzymatic activity. Consequently, the color was altered (Table 2), confirmed by comparing
the L*, a*, and b* values with those of the other treatments. The pulp and peel is a source of
phenolic compounds, which are substrates of oxidative enzymes and initiate the degradation
during the processes of cutting, fruit pulping and aeration in a finisher (Abreu et al., 2012). For
TA and TB treatment, the PPO activity was reduced by 52.49% and 39.16%, respectively.
Likewise, the POD activity was reduced by 41.74% and 33.13%, respectively. The difference of
color was 7.15 (ΔETControl TA) and 6.84 (ΔETControl TB), considerably higher than the clinically-
acceptable values reported by Rosentritt, Esch, Behr, Leibrock, and Handel (1998).
The results obtained demonstrate that ascorbic acid can reduce the PPO and POD activity,
resulting in the maintenance of the characteristic color of pitaya pulp. Herbach et al., 2006,
Leong et al., 2018 previously reported that ascorbic acid is responsible for a greater stability of
betacyanin degradation. Strack, Vogt, and Schliemann (2003) reports that a slightly acidic pH
stabilizes the betacyanins and inhibits the potential oxidation by PPOs.

the treatments (TA, TB, and Control) were analyzed using the CG-MS
technique, together with chemometric analysis. For this, the metabolic profiling was
investigated using an untargeted global metabolomics platform. In general, a total of 80
different volatile metabolites were identified in all the treatments (Table 3), which varied
according to the treatment employed. In fact, the compounds identified consisted of alcohols,
aldehydes, ketones, hydrocarbons, ether, esters, and carboxilic acids.

Obenland et al. (2016) evaluated the different varieties of pitaya and identified 34 volatile
compounds, mainly aldehydes, hydrocarbons, alcohols, ketones, esters, and furans. Célis, Gil,
and Pino (2012) reported the presence of 146 volatile compounds in another variety of pitaya
(Hylocereus megalanthus (Haw.) Britton & Rose), of which 121 of the compounds identified (83%)
consisted of terpenes, alcohols, esters, ketones, and other compounds. As shown in Table 3, 55
compounds (including 28 alcohols, 1 aldehydes, 10 ketones, 4 hydrocarbons, 9 esters, and 3
carboxilic acids) were identify in TB, followed by 50 compounds (including 13 alcohols, 8
aldehydes, 6 ketones, 10 hydrocarbons, 1 ether, 9 esters, and 3 carboxilic acids) in TA, and 47
compounds (including 18 alcohols, 7 aldehyde, 5 ketones, 7 hydrocarbons, 1 ether, 4 esters,
and 5 carboxilic acids) in the Control, and 46 compounds (57.5%) (including 14 alcohols, 8
aldehydes, 6 ketones, 10 hydrocarbons, 1 ether, 9 esters, and 3 carboxilic acids) in TA.
The volatile profile composition varied according to the pulp processing employed. As
discussed earlier, the addition of ascorbic acid (TB) decreases the level of PPO and POD
activity. Moreover, this process was useful in order to maintain almost 68% of the volatile
compounds identified in TA (pulp processing without ascorbic acid) (Table 2).
Terpenoids and furanones are formed via the metabolic pathway of carbohydrates. The
lipoxygenase (LOX), hydroperoxide lyase (HPL), and alcohol dehydrogenase (ADH) are the
main enzymes responsible for the formation of secondary alcohols, aldehydes, ketones, acids,
and esters (Schwab, Davidovich-Rikanati, & Lewinsohn, 2008). As observed in the pitaya
composition (Table 1), the levels of carbohydrate in the pulp and the acidification process can
be related to the alcohols observed in TB. Furthermore, the acidification could be related to
the conservation of its glycosylated compounds.
The PCA 3D represented 98% of the total variance (R2X [1] = 64%; R2X [2] = 32.9%; R3X
[3] = 1.1%). The data obtained were initially subjected to a PCA approach using the Pareto scale,
as shown in Fig. 1a. After PCA, PLS-DA (see Fig. 1b) was performed to identify the most similar
treatments and agglomerates. These results confirmed the data obtained from PCA, where the
volatile compounds were differently affected by the process employed.

ethylcyclopentyl, methanol (tR = 19.19 min), 10. 1,6-octadien-3-ol, 3,7-dimethyl-, (tR = 21.00 min),
11. undecane (tR = 21.01 min), 12. phenethyl alcohol (tR = 21.48 min), 13. octanoic acid, methyl
ester (tR = 22.06 min), 14. 6-tridecene (tR = 27.55 min), 15. pentadecane (tR = 30.15 min).
In addition to PCA and PLS-DA, the Variable Importance in the Projection (VIP) were
evaluated (see Fig. 1c and d). The variable importance in the projection (VIP) is a weighted sum
of squares of the PLS weight, and a VIP> 1 is generally used as a criterion to identify a
variable’s importance in the model. According to Fig. 1c, hexanal, undecane, and octanoic acid
methyl ester were the volatile compounds with the highest values of discrimination between
treatments, and were found in higher concentrations in the Control. Additionally, 1-hexanol
was observed at higher concentrations in Control and TA, indicating that the degradation
reactions are more intense in these treatments. These results reinforce the chemical
protection of the constituents of pitaya due the ascorbic acid addition.
Due to the large amount of information obtained, as well as to obtain an overview of the
metabolic variations in the different processes, heatmap analysis was performed using the
chemical markers (see Fig. 2). The 3D dendrogram (heatmap) showed that the Control had a
high degree of similarity with TA. Thus, the heatmap demonstrated that the metabolites
hexanal, undecane, octanoic acid methyl ester, hexanoic acid ethyl ester, hexanoic acid methyl
ester, 6-tridecene and pentadecane are found at a higher concentration in the Control.
Moreover, TA presented a moderate intensity of these metabolites. Concerning TB, the
presence of higher intensity metabolites belonging to the alcohols (1-butanol 3-methyl, 2,3-
butanediol phenethyl alcohol, 2,3-butanediol, 1-butanol-3-methylacetate, and trans-2-
ethylcyclopentyl-methanol), a ketone (2-hexanone) and a acid (hexanoic acid methyl ester).
Thus, the chemometric analysis was able to confirm the effectiveness of the addition of
ascorbic acid and its potential for delaying the reactions of degradation of the metabolic
pathways in processes related to the degradation of volatile compounds. These results can be
used by the industry (food, cosmetics, and others) once the effects of processing on the
chemical, physicochemical, enzymatic and volatile metabolic composition of pitaya have been
established. Thus, these results provide information about the possibilities of technological
uses of pitaya in industry processing.

4. Conclusion
Whole fruit processing with the addition of ascorbic acid (Treatment B, TB) resulted in low
polyphenol oxidase and peroxidase activity, no significant chemical or physicochemical
alterations in most parameters, and higher yields and fiber content than Treatment A (TA,
only pulp processing without ascorbic acid) or the control (whole fruit processing without
ascorbic acid). Considering the method used, the GC–MS was confirmed as an efficient
analytical technique for the separation and identification of the volatile compounds in pitaya.
Moreover, chemometric analysis proved to be fundamental for the differentiation between the
processes studied. Our results indicate that TB is a technological process which increases pulp
yield (13%) and conserves and aggregates volatile components. Due to their unique
contribution to the literature, we believe that our findings are of great importance for future
applications in the food industry.
