ABSTRACT
Essential oils from the gender Croton (Euphorbiaceae) are sources of bioactive
molecules, from which are obtained several compounds of diverse chemical
groups and great therapeutic potential in folk medicine. The essential oil of
Croton zehntneri (EOCz), popularly known as Canela-de-Cunh, possesses
anethole, estragole and eugenol as the main constituents. Based on the in vitro
antiinflammatory, antioxidant and antinociceptive properties of EOCz and/or its
main constituents, the aim of this study was to investigate the underlined
mechanism involved in the EOCz and anethole antiinflammatory action, using
models of mice paw edema of non immune and immune origin, induced by
carrageenin and ovalbumin (in desensitized animals), respectively. The animals
subchronic treatment with EOCz and anethole was also performed in order to
evaluate possible systemic toxicity. EOCz (10, 30, 100 mg/kg; p.o.) and
anethole (3, 10, 30 mg/kg; p.o.) inhibited the paw edema induced by
carrageenin about 40 and 31%, respectively. The investigation of the antiinflammatory mechanism showed that EOCz (10mg/Kg) inhibited the edema
induced by bradykinin (57%), histamine (54%), sodium nitroprusside (45%) and
substance P (33%), but did not alter those induced by serotonin, compound
48/80, arachidonic acid or prostaglandin E2. Anethole inhibited the edema
induced by bradykinin (59%), serotonin (45%), substance P (36%) and
histamine (30%) but did not interfere with the edema induced by sodium
nitroprusside. The EOCz antiedematogenic effect (35%) was also observed in a
model of immune inflammation. In summary, the present results show that
EOCz and anethole present antiedematogenic activity in a model of acute
inflammation, whose mechanism involves mainly their inhibitory action towards
kinins, but also upon the late phase mediator nitric oxide. EOCz and anethole
do not present systemic toxicity, being this important feature useful for future
phytotherapic application.
