RESUMO
Introdução: A hipertermia é um tipo de tratamento contra o câncer que envolve a
indução de calor no tumor para promover morte celular e indução do sistema
imunológico. A hipertermia por indução magnética mediada por nanopartículas
magnéticas (NPM) é baseada no aumento de calor no local do tumor decorrente da
agitação das NPM em campo magnético alternado. A técnica é considerada eficiente
e não invasiva no tratamento do câncer, pois promove a ocorrência de morte celular
no tecido tumoral por meio da hipertermia e indução do sistema imunológico.
Objetivos: Este estudo buscou determinar se a hipertermia mediada por
nanopartículas magnéticas causa aumento da taxa de necrose ou apoptose no
tumor Ehrlich induzido em camundongos, utilizando a técnica de varredura
histológica para uma melhor análise quantitativa através do software IMAGE J.
Buscou-se também inferir se houve lesões celulares, a partir da análise dos tecidos
em microscópio óptico, nos órgãos renal, esplênico, hepático e pulmonar após a
hipertermia mediada por nanopartículas magnéticas, a fim de avaliar a capacidade
de efeitos tóxicos, visíveis a microscópio óptico. Metodologia: Foram utilizados
camundongos albinos Swiss, com 6 a 8 semanas de vida. Após 20 dias da indução
do tumor de Ehrlich, foi injetado a NPM BSA - IR 780 – MnFe2O4 diretamente no
tumor de Ehrlich. Os camundongos foram submetidos à hipertermia magnética em
campo magnético alternado nos tempos zero (logo após a injeção de NPM), 24 e 72
horas, em seguida, os animais foram sacrificados e retirados o tumor e tecidos para
análise histopatológica. Resultados: A taxa de morte celular foi avaliada por meio
da análise de varredura histológica e mostrou uma maior área de morte celular no
grupo de camundongos tratados nos tempos 24 e 72 horas após a hipertermia. Os
tecidos rins, fígado, baço e pulmões dos camundongos tratados por hipertermia
foram analisados para verificação de lesões celulares, onde foi observado que as
NPM não induziram alterações histológicas visíveis por meio de microscópio óptico.
Conclusão: Este estudo determinou que houve diminuição do tumor Ehrlich de
camundongos, através da quantificação da região de morte celular no tumor, após
hipertermia mediada por nanopartículas. Não foram visualizados lesões celulares
que inferem efeitos tóxicos nos tecidos renal, esplênico, hepático e pulmonar.

ABSTRACT
Introduction: Hyperthermia is a type of cancer treatment that involves inducing heat
in the tumor to promote cell death and induction of the immune system. Magnetic
nanoparticle-mediated (NPM) magnetic induction hyperthermia is based on increased
heat at the tumor site resulting from the agitation of NPMs in alternating magnetic
field. The technique is considered efficient and noninvasive in the treatment of
cancer, as it promotes the occurrence of cell death in the tumor tissue through
hyperthermia and induction of the immune system. Objectives: This study aimed to
determine if hyperthermia causes a decrease in the Ehrlich tumor by determining
regions with cell death using the IMAGE J software, through the analysis of
histopathological scanning after hyperthermia mediated by magnetic nanoparticles. It
was also sought to infer if the cellular lesions found after hyperthermia can lead to
toxic effects on the tissues of the kidneys, spleen, liver and lungs. Method: Swiss
albino mice, 6 to 8 weeks old, were used. After 20 days of Ehrlich tumor induction,
NPM BSA - IR 780 - MnFe 2 O 4 was injected directly into the Ehrlich tumor. The
mice were submitted to magnetic hyperthermia in alternating magnetic field at zero
times (shortly after the injection of NPM), 24 and 72 hours, then the animals were
sacrificed and the tumor and tissue removed for histopathological analysis. Results:
The presence of NPM in tumors at 24 and 72 hours was determined in the tumor
sections by the Perls technique. The rate of cell death region was assessed by
histological scanning analysis and showed a greater area of cell death in the group of
mice treated at times 24 and 72 hours after hyperthermia. The tissues of the kidneys,
liver, spleen and lungs of hyperthermia treated mice were analyzed for cellular
lesions, where it was observed that the NPM did not induce visible histological
alterations by means of an electron microscope. Conclusion: This study determined
that there was a decrease in the Ehrlich tumor of mice, by quantifying the region of
tumor cell death after hyperthermia mediated by nanoparticles. Cell lesions that infer
toxic effects on renal, splenic, hepatic and pulmonary tissues were not visualized in
an electronic microscope.

Figura 1 – Na imagem podemos ver a hipertermia originada de fonte externa e interna. Na parte
esquerda, temos a utilização da hipertermia, no tumor, de fonte externa o aquecimento ocorre
na superfície do corpo, onde a temperatura irá diminuit à medida que aprofunda os tecidos,
levando a geração de calor em regiões não específicas e dissipação da maior parte de energia
térmica para tecidos saudáveis situados ao longo do caminho pelo calor induzido. Na parte
direita da imagem, temos um tumor carregado com nanopartículas magnéticas onde, através
do aquecimento destas partículas, podem direcionar o aquecimento de forma a garantir
menores danos aos tecidos laterais (normais).

submetidos a três banhos em xilol durante 1 minuto cada e em seguida foi feito
a montagem das lâminas com adição de lamínula fixada ao corte por Entellan.
Após a coloração, os tecidos foram fotografados para análise em
Microscópio de Captura Leica do laboratório multiusuário do Instituto de
Patologia Tropical e Saúde Pública.
