RESUMO
ABSTRACT
Over the years, advances in embryos in vitro production have provided efficiency in
the herds genetic improvement. Biotechnology innovation in the commercial
agricultural market allowed a single female of high zootechnical value or cases of
females not to reproduce to produce several descendants throughout their
reproductive life, reducing the intervals between generations. The process of in vitro
embryo production involves steps that include oocyte uptake, maturation, fertilization
and culturing. Brazil continues to lead and is a benchmark for the technique, but
there are still some obstacles that are related to the high cost of production especially
due to the need of a laboratory infrastructure and the variability between the results
obtained, among other challenges. What justifies the approach of biotechnology is

the importance of emphasizing that Brazil has leadership of the rapid genetic
improvement favoring the profit for the meat and milk industries. The
commercialization of in vitro embryos in large scale favors viable costs and benefits
that make the technique accessible, and it is of great importance the understanding
of the requirements that this production has to offer us. Therefore, the aim of this
review is to perform a retrospective of the main stages of the in vitro production of
bovine embryos showing the benefits that the technique possesses and the
challenges that it faces.
1 INTRODUÇÃO
Existem diversas técnicas que podem ser usadas para a coleta de oócitos
para PIV. Os primeiros procedimentos realizados eram feitos através de laparotomia

ventral média, após superovulação in vivo ou pela coleta de ovários de animais post
mortem pelo método de punção folicular, slicieng e/ou pela curetagem da parede
folicular (LAMBERT et al., 1983; VARAGO et al., 2008; WANI et al, 2000). Com o
passar dos anos, no entanto, avanços consideráveis foram obtidos que facilitaram os
meios de obtenção de oócitos em animais in vivo, popularmente conhecida como a
técnica de aspiração folicular transvaginal guiada por ultrassonografia – Ovum pick
up (OPU), que favorece os programas de melhoramento genético (ALVES et al.,
2001).
A OPU foi elaborada para satisfazer a demanda por um procedimento para
coleta de complexo cumulus-oócito, menos traumática que as abordagens cirúrgicas
utilizadas antigamente. Esta técnica possui vários benefícios e por ser pouca
invasiva, pode ser usada em qualquer fase do ciclo estral. Obtendo-se também
oócitos de fêmeas a partir de 6 meses de idade, fêmeas idosas, de animais pré-
púberes ou de animais que apresentam gestação inicial até o terceiro mês e é
possível fazer coleta em animais que estejam entre segunda e terceira semanas

após parto (GALLI et al., 1996; VIANA; BOLS, 2005). Outro benefício da técnica é a
não interferência no estado fisiológico do animal, não exige estimulação hormonal
exógena e as sessões de aspiração folicular podem ser realizadas a cada 15 dias ou
mensalmente (GALLI; LAZZARI, 2000; NAGAI, 2000; VARAGO et al., 2008). Essa
periodicidade dos procedimentos até mesmo mensalmente, possui em torno de 40%
de índices de produção em relação ao seu rendimento, podendo superar os índices
obtidos pela transferência de embriões, tornando esta técnica rentável, relacionando
o custo ao benefício (VARAGO et al.,2008). Além disso a técnica de OPU possibilita
a coleta em animais de alto valor genético que por algum motivo possuem
infertilidade, tornando-se possível a obtenção de oócitos gerando produtos
descendentes (BOLS et al., 2012).
A coleta de oócitos em fêmeas bovinas é realizada pela punção folicular com
uma agulha acoplada a uma sonda transvaginal, de forma que os folículos a serem
puncionados são observados na tela do ultrassom. Os oócitos aspirados dos
folículos devem ter diâmetro de 2 a 8 mm e os folículos menores que o mínimo de 2
mm são incapazes de recomeçar a fase de meiose e os maiores de 8 mm
normalmente se deparam em estágio de atresia para a maturação, tornando essas
duas formas inviáveis (GONÇALVES et al., 2008).
