ESQUELÉTICO DE RATOS SÉPTICOS SEDENTÁRIOS E TREINADOS

Resumo
Sepse é a resposta inflamatória sistêmica secundária a um processo infeccioso de alta incidência
e elevada mortalidade. A miopatia esquelética, causada pela sepse, gera atrofia muscular e
fraqueza generalizada que estão relacionadas a disfunção mitocondrial, produção exacerbada de
espécies reativas de oxigênio (ERO) e aumento da degradação protéica. A hipótese que será
testada nesse projeto é que no modelo de sepse grave, há aumento da atividade do sistema
proteolítico ubiquitina proteassoma (UP) no músculo esquelético, promovendo atrofia e fraqueza
muscular, e que o condicionamento físico prévio a sepse poderia amenizar os efeitos dessa
enfermidade sobre a miopatia esquelética . O objetivo será avaliar a atividade do sistema
proteolítico UP no músculo equelético em modelo experimental de sepse grave e os efeitos do
treinamento físico (TF) prévio sobre esse sistema de degradação. Os experimentos serão
realizados em ratos Wistar, inicialmente distribuídos em dois grupos: Não-Treinado e Treinado. O
grupo Treinado será submetido a protocolo de TF, de predominância aeróbica, com duração de
oito semanas, em esteira rolante (60 minutos, 5 dias por semana, a 60% da velocidade máxima
encontrada em teste de esforço máximo). Ao final do protocolo, o grupo Não-Treinado será
dividido em grupo Sham (falsa cirurgia) e CLP (indução da sepse por ligação e punção cecal); e o
grupo Treinado da mesma maneira: ShamT e CLPT, totalizando quatro grupos experimentais. Os
quatro grupos serão submetidos aos procedimentos cirúrgicos e após dois dias sacrificados
sendo coletado sangue e retirados os músculos: sóleo, plantar e diafragma para as devidas
análises. A atividade do sistema de degradação protéica UP será avaliada através da expressão
gênica e da expressão protéica das E3 ligases MURF-1, Atrogin-1 e E3a, e a deubiquitinase USP
28. A expressão gênica será avaliada por RT- PCR utilizando primers específicos e Sybr Green
Master Mix. A expressão protéica será avaliada através da técnica de Western Blotting utilizando
anticorpos primários e secundários.
