RESUMO
Nesse trabalho, são apresentados aspectos relacionados ao desenvolvimento das principais
classes farmacológicas utilizadas no tratamento de hipercolesterolemia, explorando-se as
características estruturais importantes a sua ação biológica.
ABSTRACT
In this work, the development of main pharmacological classes used in the treatment of
hypercholesterolemia is described, exploring the most important structural features to their
biological activity.
1 INTRODUÇÃO
A hipercolesterolemia é um importante fator de risco para o desenvolvimento de
doenças cardíacas. Isso porque o excesso de colesterol pode levar ao desenvolvimento de placas
de ateroma no endotélio das importantes artérias do nosso organismo, podendo contribuir para

Após extensas modificações estruturais, o composto SCH 48461 foi identificado com o
melhor perfil hipocolesterolêmico entre os estudados ao reduzir o nível de CE no fígado em
93% a 10 mg/Kg, uma porcentagem superior a qualquer outra substância testada até então. Além
disso, ele também conseguiu diminuir os níveis de CS em 43% na mesma concentração em
ensaios animais. Vale ressaltar que o valor de IC50 encontrado para essa azetidinona contra
ACAT foi de 26 µM, reforçando divergências entre os resultados de testes in vitro e in vivo,
