Resumo
Os ésteres de açúcar são compostos de alta relevância, com aplicação nas indústrias
farmacêuticas e de alimentos. O processo químico de produção destes ésteres é
complicado devido à grande quantidade de grupos hidroxila com reatividades químicas
semelhantes presentes nos carboidratos e à necessidade de modificação regiosseletiva
de carboidratos para se obter o composto desejado; na síntese química, há necessidade
de adição de grupos protetores das hidroxilas que não se deseja esterificar e que devem
ser removidos para obtenção do produto final. O processo enzimático é vantajoso, uma
vez que enzimas são e podem produzir o produto desejado em uma única etapa de
reação. Outro problema na produção de ésteres de açúcar é a utilização de solventes
orgânicos, como dimetilformamida e piridina, que são substâncias tóxicas e podem
resultar na formação de compostos secundários indesejáveis, trazendo diversos
problemas ambientais. Com o avanço da química verde, foram desenvolvidos solventes
ambientalmente mais corretos para a síntese de ésteres, como os solventes eutéticos
profundos (DESs), que vêm demonstrando resultados promissores. O objetivo deste
trabalho é a síntese enzimática de ésteres de açúcar, utilizando solventes eutéticos
naturais (NADESs), puros ou em misturas, produzidos com açúcares e cloreto de colina.
Como catalisadores da reação serão utilizados o sólido fermentado de Burkholderia
contaminans LTEB11 contendo lipases, a lipase LipC12 imobilizada e, para
comparação, a lipase B de Candida antarctica (CALB). Os resultados deste trabalho
poderão permitir a síntese de ésteres de açucares em meios reacionais considerados
“verdes” como alternativa aos solventes orgânicos atualmente utilizados.
Ésteres de ácidos graxos de açúcar, também conhecidos como ésteres de açúcar (EAs),
são biossurfactantes não iônicos capazes de reduzir a tensão superficial e promover a
mistura de componentes imiscíveis, como óleo e água (Ren & Lamsal, 2017). Essa
propriedade se deve à natureza anfifílica da molécula, uma vez que os EAs são
glicoconjugados formados a partir de ligações covalentes entre um carboidrato e um
lipídio, contendo uma fração hidrofílica e uma fração hidrofóbica (Villalobos et al., 2018;
Ibinga et al., 2019).
Por outro lado, a rota enzimática é um processo que não requer a proteção ou
desproteção dos grupos hidroxila (Adachi & Kobayashi, 2005), uma vez que enzimas
são regiosseletivas. Além disso, a rota enzimática exige baixa temperatura de reação,
permite a utilização de solventes mais ecológicos, além de propiciarem altos
rendimentos e maior pureza do produto de interesse (Neta et al., 2012; Siebenhaller et
al., 2018). As lipases (EC 3.1.1.3), são as mais indicadas para realizar reações de
transesterificação para a síntese de ésteres de açúcar devido à sua eficiência e
versatilidade em diferentes condições (Gumel et al., 2011).
O objetivo do trabalho é a síntese enzimática de ésteres de açúcar, utilizando solventes
eutéticos profundos naturais também conhecidos pela sigla em inglês NADES (Natural
Deep Eutectic Solventes) puros ou misturados, produzidos com cloreto de colina e
açúcares. Serão utilizadas lipases naturalmente imobilizadas no sólido fermentado,
provenientes do fermentado em estado sólido de Burkholderia contaminans LTEB11
(SFBC), a lipase LipC12 imobilizada e, para fins comparativos, a lipase B de Candida
antarctica (CALB). Os resultados possibilitarão a produção de ésteres de açúcar em
meios reacionais mais ecológicos como alternativa aos solventes orgânicos tradicionais.
2 REVISÃO DA LITERATURA
Uma categoria especial de DES são os solventes eutéticos profundos naturais (em
inglês, natural deep eutectic solvents, NADES), que são formulações compostas por
matérias-primas renováveis derivadas de metabólitos vegetais, tais como ácidos graxos,
amidos, açúcares e ácidos orgânicos (Jeliński et al., 2019). NADES têm sido objeto de
crescente investigação devido às suas características intrínsecas, tais como a
sustentabilidade e a ausência de riscos à saúde, o que os torna escolhas altamente
promissoras para diversas aplicações industriais (Lapeña et al., 2019; Xia et al., 2020)

5 ESTRATÉGIA DE AÇÃO
Reações de síntese: serão realizadas reações de síntese com cada uma das
enzimas (LipC12, SFBC e CALB) em diferentes concentrações. Em um frasco
Erlenmeyer com tampa serão adicionados o de DES e o (217 mmol) de acetato de vinila.
Para fins de comparação será realizado a reação em meio livre de solvente onde será
adicionada a D-glucose e o acetato de vinila. Após essa etapa, os frascos Erlenmeyer
serão colocados em um agitador orbital e incubados por 24 horas a 250 rpm e
temperaturas de 37 a 60 °C. Após o período de incubação, a enzima será adicionada
ao frasco e o sistema será incubado novamente sob as mesmas condições por mais 72
horas. Será coletada uma alíquota de 500 µL dos meios de reação para posterior análise
por cromatografia líquida de alta eficiência (CLAE), ressonância magnética nuclear
(RMN) e espectroscopia de infravermelho (IR).
