RESUMO
Durante milhes de anos convivendo no mesmo ambiente, plantas e patgenos
desenvolveram um complexo sistema de interao entre eles. Patgenos utilizam um
grande nmero de estratgias para superar a barreiras de defesas e iniciarem a
colonizao nos tecidos do hospedeiro. Enquanto isso, as plantas apresentam um
complexo sistema de defesa para impedir a invaso dos patgenos e, em consequncia,
o desenvolvimento de doena. Esta batalha resulta em uma constante presso de
seleo entre os organismos envolvidos. Neste captulo so apresentados os principais
componentes envolvidos na interao planta-patgeno, que vo desde a ativao de
defesa desencadeada por reconhecimento de padres moleculares associados aos
patgenos (PAMPs), o papel fundamental de efetores e protenas de resistncia para o
desenvolvimento de doenas, as ferramentas de bioinformtica utilizadas para
predio de candidatos a genes efetores no genoma dos patgenos e o potencial uso
dos efetores na proteo da agricultura e ecossistemas naturais.
SUMMARY
Millions of years coexisting in the same environment developed a complex
interactions system between plants and pathogens. A large number of strategies are
used by pathogens to break the plant defense barriers in order to invade and colonize
plant tissues. At the same time a profusion of other strategies are used by plants to
avoid or minimize the pathogen invasion and development. As a consequence of this
battle there is a constant selection pressure in both organisms involved, resulting in a
strategy game which will never have a final winner. Here we reviewed the main
components involved in plant pathogen interaction from the Pathogen-Associated
Molecular Pattern (PAMP) and their recognition by PAMP receptors activating
defense, the role of effectors and R proteins and their importance to help pathogens
and plants respectively in this eternal war for survival, the bioinformatic tools to
predict putative effectors and its potencial use to protect crops and natural ecosystems.
INTRODUO
Milhes de anos de convvio no mesmo ambiente favoreceu o desenvolvendo
de complexo sistema de comunicao entre plantas e microrganismos. Para toda
interao planta-microrganismo, a troca de sinais representa a primeira e fundamental
etapa. Assim, a habilidade de perceber e responder a sinais do ambiente
fundamental para a sobrevivncia (Tyler et al., 2006).
Patgenos apresentam a habilidade de reconhecer sinais das plantas para
iniciar a infeco (Dalio et al., 2014). Por sua vez, as plantas esto expostas
constantemente a mudanas no ambiente, ataque de pragas e doenas. Diferentemente
dos animais, plantas no apresentam um sistema imune adaptativo e nem clulas de
defesa mveis, como os macrfagos (Chisholm et al., 2006), no entanto
desenvolveram um sistema imune simples, porm eficiente, que ativado aps o
reconhecimento de sinais da presena do microrganismo (Gassmann & Bhattacharjee,
2012; Dalio, 2013).
Os processos de reconhecimento pelo sistema imune de plantas podem ser
dividido em duas frentes: (i) aqueles associados imunidade desencadeada por
PAMPs ou MAMPs (pathogen or microbe associated molecular patterns), e (ii)
aqueles associados imunidade desencadeada por efetores. O primeiro usualmente
denominado PTI (PAMP-triggered immunity) e o segundo ETI (effector-triggered
immunity) (Jones & Dangl, 2006). Os PAMPs ou MAMPs, so reconhecidos por
receptores PRRs (PAMP-recognition receptor) localizados na superfcie da
membrana celular ou interior da clula, que desencadeiam uma cascata de transduo
de sinal ativando a resposta de defesa (Hogenhout et al., 2009). Este tipo de sistema
de defesa permite que as plantas respondam rpida e eficientemente a uma gama
ampla de patgenos (Roux et al., 2014).
Patgenos adaptados so capazes de secretar um arsenal de protenas que
podem suprimir PTI, resultando em susceptibilidade desencadeada por efetores (ETS,
effector-triggered susceptibility) (Birch et al., 2009).

Estudos envolvendo bactrias Gram-negativas com sistema de secreo tipo
III (T3SS) revelaram que estas bactrias secretam protenas no interior das clulas que
desencadeiam reaes de hipersensibilidade. Essas molculas foram portanto
chamadas de fatores de avirulncia. No entanto, as mesmas protenas cunhadas como
fatores de avirulncia foram encontradas contribuindo para a virulncia em relaes de
susceptibilidade. Deste modo, havia um impasse no uso do termo avirulncia porque
ele no era correto em todos os casos, demonstrando a necessidade de um termo
neutro. Da surgiu o termo efetores resolvendo o impasse e vem sendo aceito e
utilizado na fitopatologia.
Alguns autores ainda insistem em definir efetores como molculas que
suprimem a defesa das plantas. Como j mencionado acima, este tipo de definio no
correta porque alguns efetores apresentam dualidades em sua funo dependendo do
hospedeiro, ou seja, pode ser relacionado tanto a patogenicidade ou ativao de
resistncia. Por muito tempo a definio mais aceita, proposta por Kamoun, 2009 era:
Efetores, molculas secretadas por um microrganismo que alteram a estrutura/funo
do hospedeiro. Esta definio perdeu um pouco de espao porque nem todos os
efetores so secretados e nem sempre o organismo alvo um hospedeiro. Nesse
como
efetores
apoplsticos,
os
efetores
intracelulares
ou
Reprogramao transcricional,
da
interao
direta,
altamente
especfica,
entre
as
protenas
hospedeiro/patgeno de acordo com a teoria gene a gene (Flor, 1971; Boller & Felix,
2009). Contudo, os efetores podem ser reconhecidos de forma indireta, conhecida
como teoria guarda (Flor, 1971; Jones & Takemoto, 2004). Nesse reconhecimento
indireto, as protenas R reconhecem efetores ligados a protenas da planta chamadas
alvos de efetores de patgenos representados na Figura 4 como alvo dos efetores
citoplasmticos (AEC). A maior evidncia dessa teoria guarda foi encontrada no
sistema R/Avr entre Arabidopsis thaliana e Pseudomonas syringae pv. Tomato, onde
RIN4 modificada pela Avr dessa bactria e essa modificao ativa a protena R
(RPM1) resultando na resistncia dessa planta (MACKEY et al. 2002).

Diferentes patgenos como bactrias, fungos, oomicetos e vrus que produzem
efetores para atacar diferentes plantas, que por sua vez, usam seus genes de resistncia
para evitar a doena (Tabela 2).

gerado pelo sequenciamento Illumina, 700 pb contra de 50 a 300 pb, conferindo uma
maior acurcia dos dados, 99,9% contra 98% obtidos no sequenciamento Illumina. O
pirosequenciamento gera um menor nmero de dados, cerca de um milho de
fragmentos, enquanto o Illumina gera mais de 3 milhes de fragmentos. Isto torna o
sequenciamento Illumina mais demorado, de um a 10 dias, dependendo do tamanho
do fragmento gerado; em contrapartida, o pirosequenciamento pode ser realizado em
24 horas. Em termos de custo, o sequenciamento Illumina mais vantajoso: US$ 0,05
a 0,15 por milho de bases, enquanto que, no pirosequenciamento o custo de
US$ 10 (LIU et al. 2008; QUAIL et al. 2012). Outro aspecto vantajoso de ambas as
plataformas a sua aplicao no s no sequenciamento de genomas mas tambm no
sequenciamento de transcriptoma do organismo, metodologia esta denominada
RNAseq.
O emprego desta tcnica, tanto na genmica como na transcriptmica, tem
gerado muitos trabalhos com fitopatgenos e a sua interao com o hospedeiro,
visando principalmente, a busca por molculas efetoras e a resposta adaptativa da
planta a estas molculas, evidenciando a coevoluo deste sistema. Como exemplo
desta abordagem, podemos citar a descoberta de um gene de virulncia envolvido na
induo de tumor em uma planta lenhosa (RODRGUEZ-PALENZUELA et al. 2010);
microevoluo e relao filogentica de efetores P. syringae na adaptao ao
hospedeiro (CAI et al. 2011); anlise de polimorfismo das protenas candidatas a
efetoras secretadas pelo haustrio de Puccinia striiformis f. sp. tritici (CANTU et al.
2013); a identificao de genes candidatos a efetores no transcriptoma do nematoide
do n da raiz do arroz Meloidogyne graminicola (HAEGEMAN et al. 2013); busca por
genes efetores expressos na interao com o hospedeiro, no transcriptoma de
Melampsora lini (NEMRI et al. 2014); comparao entre genomas da mesma espcie a
qual pertence o patgeno, avaliando a conservao das protenas efetoras ao longo do
tempo evolutivo (QI et al. 2011); e a identificao de efetores de domnio
caracterizado, como os RXLR, no genoma de patgenos de grande impacto na
economia agrcola (THAKUR et al. 2013).
Para que estas diversas abordagens tivessem sucesso, simultaneamente ao
aparecimento do sequenciamento de nova gerao, diversas ferramentas de
bioinformtica foram desenvolvidas, visando analisar o secretoma dos patgenos
permitindo a predio de protenas efetoras e a minerao destas a partir de domnios
