3 – Material e métodos
3.1 Obtenção de cepas knockouts: Estabelecer uma metodologia para padronizar a tecnologia
CRISPR-Cas9 para silenciar os genes sps (spsD, spsL, spsO, spsP e spsQ), genes do operon ica
(icaA, icaD e icaC) e genes do sistema agr (agrA, agrB, agrD e agrC). A partir de uma cepa
de referência (ED99) (TAGLIAFERRI et al., 2019). A técnica já está bem padronizada para
outros grupos bacterianos, inclusive outros Gram-positivos.
3.2 Formação de biofilme em dispositivos médicos: As amostras serão testadas quanto a
capacidade de formação de biofilme em dispositivos médicos (material ortopédico de titânio,
fios de sutura, cateter, sondas e drenos). A comparação será feita com uma cepa de referência
(ED99) e duas amostras selvagens (uma sensível à meticilina e outra resistente), já
caracterizadas previamente.
- Avaliação da estrutura e arquitetura do biofilme através da microscopia eletrônica de
varredura e confocal (POMPILIO et al., 2015).
- Avaliação da viabilidade do biofilme através da contagem de UFC (MORRISON et al.,
2016).
- Avaliação quantitativa do biofilme através do ensaio colorimétrico (STEPANOVIC et al.,
2007).
- Avaliação da composição do biofilme através da metodologia de Chiba e colaboradores
(2015).
3.3 Modelo de infecção por Galleria mellonella: Avaliar a virulência das amostras silenciadas
através do ensaio in vivo, com adaptações, em Galleria mellonella. (CLAVIJO-GIRALDO et
al., 2016)
